分泌性中耳炎患者耳积液中miR-146a和miR-155的表达研究 ?(1)

     【摘要】 目的 探讨分泌性中耳炎(SOM)患者耳积液中微小核糖核酸-146a(miR-146a)和miR-155的表达及临床意义。方法 58例SOM患者作为研究对象(SOM组),另选取30名体检健康者作为对照组。应用实时荧光定量PCR法检测2组血浆及SOM组耳积液miR-146a和miR-155表达情况,应用ELISA检测2组血浆及SOM组耳积液IL-2、IFN-γ、IL-4、IL-10水平,并对miR-146a、miR-155与辅助性T淋巴细胞1(Th1)、Th2细胞比例以及Th1/Th2进行相关性分析。结果 与对照组相比,SOM组血浆、耳积液中miR-146a、miR-155、CD4+ Th1、CD4+ Th2、Th1/Th2、IL-2、IFN-γ、IL-4、IL-10、IFN-γ/IL-4均升高(P均< 0.05);与SOM组血浆值相比,SOM组耳积液中miR-146a、miR-155、CD4+ Th1、CD4+ Th2、Th1/Th2、IL-2、IFN-γ、IL-4、IL-10、IFN-γ/IL-4表达水平均升高(P均< 0.05);Pearson相关分析显示:SOM组患者耳积液miR-146a、miR-155水平与CD4+ Th1细胞比例、CD4+ Th2细胞比例、Th1/Th2均呈正相关,耳积液miR-146a、miR-155表达与IL-2、IFN-γ、IL-4、IL-10呈正相关。结论 SOM患者耳积液中miR-146a和miR-155高表达,miR-146a、miR-155可能通过影响Th1/Th2平衡而促进炎症进展。

    【Abstract】 Objective To investigate the expression levels and clinical significance of microRNA-146a (miR-146a) and microRNA-155 (miR-155) in the ear effusion of patients with secretory otitis media (SOM). Methods A total of 58 SOM patients were assigned into the SOM group, and 30 healthy subjects were recruited into the control group. The expression levels of miR-146a and miR-155 in the plasma between two groups and in the auricular effusion in the SOM group were quantitatively measured by real-time fluorescence quantitative PCR. The expression levels of interleukin-2 (IL-2), interferon-γ(IFN-γ), interleukin-4 (IL-4) and interleukin-10 (IL-10) in the plasma between two groups and in the auricular effusion in the SOM group were measured by enzyme-linked immunosorbent assay (ELISA). The correlation between miR-146a, miR-155, Th1 and Th2 proportion and Th1/Th2 was analyzed. Results Compared with the control group, the expression levels of miR-146a, miR-155, CD4+ Th1, CD4+ Th1, Th1/Th2, IL-2, IFN-γ, IL-4, IL-10, IFN-γ/IL-4 were significantly up-regulated in the SOM group (all P < 0.05). Compared with plasma expression of the SOM group, the expression levels of miR-146a, miR-155, CD4+ Th1, CD4+ Th2, Th1/Th2, IL-2, IFN-γ, IL-4, IL-10, IFN-γ/IL-4 in the auricular effusion were up-regulated (all P < 0.05).Pearson’s correlation analysis demonstrated that the expression levels of miR-146a and miR-155 were positively correlated with the proportion of CD4+ Th1, CD4+ Th2 and Th1/Th2. The expression levels of miR-146a and miR-155 were positively correlated with IL-2, IFN-γ, IL-4 and IL-10 in the auricular effusion. Conclusions The miR-146a and miR-155 are highly expressed in the ear effusion of SOM patients. The miR-146a and miR-155 can promote the progression of inflammation probably by affecting the balance of Th1/Th2., 百拇医药(王庆亮 徐锦)
