禹州漏芦总黄酮抑制佐剂性关节炎大鼠Wnt信号(1)
[摘要]研究地道药材禹州漏芦主要活性成分总黄酮(FELT)对佐剂性关节炎(AA)大鼠Wnt信号的影响。采用大鼠关节炎评分法和MTT检测FELT对AA大鼠的治疗作用,Real-time qPCR检测FELT灌胃治疗对AA大鼠滑膜中Wnt信号关键基因β-catenin,C-myc和cyclin D1表达的影响,Real-time qPCR检测FELT灌胃治疗对AA大鼠滑膜中Wnt信号上游负调控基因SFRP 1,2,4,5表达的影响。结果发现FELT对AA大鼠关节炎评分和MTT值有显著的抑制作用,对Wnt信号关键基因β-catenin,C-myc和cyclin D1表达有显著的抑制作用,对SFRP 1,2,4表达有显著的上调作用。FELT灌胃治疗对AA大鼠有较好的治疗作用,这种治疗作用可能是通过对Wnt信号的抑制实现的。

[Abstract]The role of flavonoids of Echinps latifolius(FELT) in Wnt signaling was investigated in adjuvant arthritis (AA) rats. The therapeutic effects of FELT on AA rats were detected by rat arthritis score and MTT. The effect of FELT gavage treatment on the Wnt signaling key gene β-catenin, C-myc and cyclin D1 in synovium from AA rats was detected by Real-time qPCR, and the effects of FELT gavage treatment on the upstream negative regulation gene SFRP 1,2,4,5 in synovium from AA rats were detected by Real-time qPCR. The results showed that FELT gavage treatment significantly inhibited arthritis score and MTT values in AA rats, significantly inhibited the expression of the Wnt signaling gene β-catenin, C-myc and cyclin D1, significantly up-regulated the expression of the upstream negative regulation gene SFRP 1,2,4. FELT has a better therapeutic effect for AA rats.

类风湿性关节炎(rheumatoidarthritis,RA)是一种慢性、系统性、多关节性疾病,发病机制仍不清楚,世界范围内RA发病率为0.5%~1.0%,部分地区高达5%[1-2]。成纤维细胞样滑膜细胞(fibroblast-like synoviocytes,FLS)在RA发病机制中发挥重要作用[3-4]。Wnt信号参与了RA的发病机制,与FLS异常活化增殖、持续的滑膜炎症、软骨侵蚀、骨的破坏等有关,Wnt通路相关分子可能是潜在的RA治疗靶点[5]。禹州漏芦为菊科华东蓝刺头Echinps latifolius Tausch的干燥根,主产于安徽、湖北、河南等省,其中以安徽、湖北的产量最大、质量最好。禹州漏芦具有抗炎、镇痛、耐缺氧、抗疲劳、降血压及降血脂等药理作用,是一种极具研究开发价值的中草药[6]。本文采用滁州产禹州漏芦为研究对照,分离提取其总黄酮,采用佐剂性关节炎大鼠为AA,大鼠关节炎评分法和MTT检测禹州漏芦总黄酮(FELT)对AA大鼠的治疗作用,Real-time qPCR检测FELT灌胃治疗对AA大鼠Wnt信号和对Wnt信号上游负调控基因SFRP 1,2,4,5表达的影响,为禹州漏芦的综合开发积累实验基础,为RA治疗提供新思路。, 百拇医药(缪成贵 徐健 高虎 刘亮亮 周国梁 秦梅颂 陈建中 李成凤)
