痰热清注射液对肺癌患者外周血淋巴细胞免疫活性的影响(1)
[摘要]为探讨痰热清注射液对肺癌患者淋巴细胞免疫活性的影响，本研究采用密度梯度法分离晚期肺癌患者或健康人外周血淋巴细胞用于后续实验，以健康人外周血淋巴细胞作为阳性对照。通过MTT法检测痰热清注射液对植物血凝素(PHA)刺激淋巴细胞增殖能力的影响，通过ELISA法检测痰热清注射液对淋巴细胞分泌IFN-γ，TNF-α水平的影响，通过流式细胞术(flowcytometry，FCM)检测单独或与不同浓度痰热清注射液共培养后的淋巴细胞各亚群比例，通过LDH释放法检测各实验组细胞毒性T细胞(cytotoxicTcells，CTL)及NK细胞杀伤活性。结果表明，肺癌患者外周血淋巴细胞各项免疫活性指标均较正常人明显下降，而与痰热清注射液共培养的淋巴细胞，其增殖活性及分泌IFN-γ及TNF-α的能力均有所提高(P<0.05)。与痰热清注射液共培养的淋巴细胞各亚群中，CD3/1，CD3/1CD4/1，CD3-CD16+56/1细胞比例均明显增高，而CD4/1CD25/1调节性T细胞比例明显下降(P<0.05)。同时痰热清注射液可明显促进CTL细胞及NK细胞的杀伤活性(P<0.05)。综上所述，痰热清注射液对肺癌患者淋巴细胞的免疫活性有明显的促进作用，并对肺癌机体抗肿瘤免疫力有改善作用。
