黄蜀葵花中黄酮类成分对前脂肪细胞增殖、分化及胰岛素抵抗的影响(1)
[摘要] 黃蜀葵花中富含黄酮类成分,具有保护心血管、改善肾功能等作用。该文采用3T3-L1前脂肪细胞诱导分化的成熟脂肪细胞,建立胰岛素抵抗模型,分为正常组,模型组,异槲皮苷(HY)、金丝桃苷(JY)、槲皮素(QT)、槲皮素-3′-O-葡萄糖苷(QG)、棉皮素-8-O-β-葡萄糖醛酸(GG)给药组,BCA法检测细胞培养液中葡萄糖的含量,荧光定量qPCR检测PPARγ,C/EBPα,SREBP-1,脂联素,内酯素,抵抗素基因mRNA的表达水平。结果显示,5种黄酮类化合物5 μmol·L-1浓度时可促进3T3-L1前脂肪细胞的增殖,浓度为100 μmol·L-1时可抑制前脂肪细胞的增殖;与正常组比较,模型组细胞对葡萄糖摄取量降低(P<0.01);与模型组相比,5种黄酮类化合物100 μmol·L-1 给药组细胞葡萄糖消耗量显著上升(P<0.01),PPARγ,C/EBPα,脂联素表达明显增加(P<0.01),SREBP-1,抵抗素,内酯素的表达明显降低(P<0.01),促使脂肪细胞分化。研究表明,HY,JY,QT,QG,GG能调控前脂肪细胞增殖,促进脂肪细胞分化及糖脂代谢相关因子PPARγ,C/EBPα,SREBP-1,脂联素,内酯素,抵抗素的表达,促进前脂肪细胞分化,增加葡萄糖利用,从而改善胰岛素抵抗。

[Abstract] Abelmoschus manihot was rich in flavonoids,which has been reported the activity on protecting angiocarpy and improving renal function. This study aimed to explore the action mechanism of five flavonoids from A. manihot on how to ameliorating insulin resistance through the regulation of the glucose and expression of PPARγ,C/EBPα,SREBP-1,resistin,visfatin,adiponectin in 3T3-L1 adipocytes. After the 3T3-L1 preadipocytes were differentiated into mature adipocytes,insulin resistance model was built. Insulin resistance adipocytes were treated with 5,100 μmol·L-1 quercetin,isoquercitrin,hyperoside,quercitrin-3′-O-glucoside,gossypetin-8-O-β-glucoside. The glucose was indirectly determined by BCA kit. The mRNA expression levels of PPARγ,C/EBPα,SREBP-1,resistin,visfatin,adiponectin were detected by real-time quantitative PCR. Results showed that five flavonoids at 5 μmol·L-1 could accelerate preadipocytes proliferation and inhibit that at 100 μmol·L-1 Compared with the normal group,glucose uptake reduced significantly in model group (P<0.01). With the treatment of five flavonoids at 100 μmol·L-1,glucose consumption increased significantly (P<0.01). The high expression of PPARγ,C/EBPα,adiponectin expression was significantly increased (P<0.01),and low expression of SREBP-1,resistin,visfatin after respective administration with five flavonoids at 100 μmol·L-1 promoted adipocyte differentiation. This study showed that,HY,JY,QT,QG,GG can control preadipocytes proliferation,promote adipocyte differentiation and regulate the expression of relative factors with lipid metabolism,such as PPARγ,C/EBPα,SREBP-1,adiponectin,resistin,visfatin,increasing glucose utilization and improving insulin resistance in 3T3-L1 adipocyte.
