基于UPLC—Q—TOF—MS技术的芩百清肺浓缩丸血清药物化学初步研究(1)
[摘要]应用UPLC-Q-TOF-MS技术和血清药物化学理论分析检测芩百清肺浓缩丸的入血成分。灌胃给予大鼠芩百清肺浓缩丸溶液后,肝门静脉采血,血清经SPE处理,采用ACQUITY UPLC BEH C18色谱柱(2.1 mm×100 mm,1.7 μm),0.1%甲酸水(A) -0.1%甲酸乙腈(B)流动相系统进行梯度洗脱,流速0.3 mL·min-1,柱温35 ℃。通过对比分析体外样品、空白血清及含药血清,同时结合Peakview和Metabolitepilot软件鉴定和表征其血中移行成分,在给药后的大鼠血清中分析鉴定了28个血中移行成分,其中18个原型成分,10个代谢产物。应用UPLC-Q-TOF-MS技术和血清药物化学理论较为全面地阐明了芩百清肺浓缩丸在大鼠血清中的移行成分,为进一步研究其体内代谢过程以及药效物质基础提供了科学依据。

[Abstract]To analyze the main components of Qinbai Qingfei concentrated pellets in rat serum with UPLC-Q-TOF-MS technology and serum pharmacochemistry theory. After gavage administration with Qinbai Qingfei concentrated pellets, blood was collected from hepatic portal vein. ACQUITY UPLC BEH C18(2.1 mm×100 mm, 1.7 μm) was used, with 0.1% formic acid agueous solution(A)-0.1%formic acid and acetonitrile(B) as the mobile phase for gradient elution. The flow rate was 0.3 mL·min-1, the column temperature was maintained at 35 ℃. Through the comparative analysis fingerprints of Qinbai Qingfei concentrated pellets, drug containing-serum and blank serum, and with the help Peakview and Metabolitepilot software, components in serum were defined. A total of 28 compounds were identified, including 18 prototypes and 10 metabolites. As a result, UPLC-Q-TOF-MS technology and serum pharmacochemistry theory were applied to comprehensively expound Qinbai Qingfei concentrated pellets′constituents migrating to rat serum, and provide scientific basis for further studies for in vivo metabolic process and effective material base.

中藥血清药物化学是以传统药物化学方法为基础,综合应用多种现代技术,分析鉴定口服中药后血清中移行成分,研究其与药效相关性,确定中药药效物质基础的应用学科[1]。此方法自提出以来已经被成功应用于多种中药以及中药复方的研究[2-3],研究结果不仅揭示中药药效物质基础,而且为解析方剂配伍规律、阐释中药药理学特性及创新药物设计等中医药关键科学问题提供了科学有效的方法。
芩百清肺浓缩丸(以下简称芩百,发明专利证书号ZL03102049.6),是国家食品药品监督管理总局批准的第一个用于临床观察治疗小儿支原体肺炎的中药制剂,由黄芩、百部、紫菀、桔梗、麦冬、地龙6味中药组成,具有清热解毒、降气平喘、润肺止咳之功效,适用于儿童支原体肺炎的治疗。前期实验表明,其能提高免疫功能和促进气道上皮细胞的修复,能有效减轻支原体对肺部组织的炎性病变,促进上皮细胞的恢复[4-6]。然而芩百的药效物质基础尚未完全阐明,本文利用血清药物化学方法以及UPLC-Q-TOF-MS技术进一步研究芩百的血中移行成分,旨在揭示其药效物质基础。
