痰热清注射液质量关键影响因素辨识方法初探(1)
[摘要]为辨识痰热清注射液配液过程中影响最终产品质量的关键因素,采集了24个实际生产批次中5种中药提取物的近红外漫反射光谱,记录了配液过程中的31个操作参数,作为自变量;记录最终产品的总含固量和灯检合格率,作为因变量。采用偏最小二乘回归方法建立了定量校正模型,得到自变量和因变量之间的相关系数。所得总含固量和灯检合格率定量校正模型的校正相关系数分别为0911 9,0873 8,交叉验证的相关系数分别为0724 2,0795 0。根据相关系数筛选出4个与总含固量相关性较强的因素,8个与灯检合格率相关性较强的因素,并结合生产经验,对这些因素的实际意义进行了分析。该文所建方法对配液过程中影响痰热清注射液最终产品的因素进行了初步辨识,结果合理,可推广应用于解决类似问题。

[Abstract]To recognize the key influential factors during the liquid preparation process of Tanreqing injection, the near infrared(NIR) spectra of the raw materials and the operating parameters of 24 batches of physical manufacturing were recorded as independent variables, and the total soluble solids contents and the light inspection acceptance rate of the final products were collected as dependent variables The calibration models were developed using the partial leastsquare regression (PLSR) method, and the correlation coefficients between the independent variables and the dependent variables were calculated For the quantitative models, the correlation coefficients for the calibration and inner cross validation of total soluble solids contents and the light inspection acceptance rate reached 0911 9, 0724 2 and 0873 8, 0795 9, respectively Using the correlation coefficients diagrams, several key influence factors were preliminarily determined, and the physical significance were analyzed combined with production experience This work demonstrated that NIR spectroscopy with PLSR algorithm could be used for the key influential factors recognization during the liquid preparation process of Tanreqing injection and can be popularized to solve similar problems

痰熱清注射液是由熊胆粉、山羊角、黄芩、连翘、金银花5种药材制得,主治风温肺热病属痰热阻肺症,主要用于治疗急性支气管炎、慢性支气管炎急性发作以及上呼吸道感染[13],2009年卫生部公布的《甲型H1N1流感诊疗方案(2009试行版第三版)》中,痰热清注射液被列为重症与危重症辩证治疗方案治疗药物,疗效显著。痰热清注射液的生产过程中,先将以上5种药材分别进行提取或水解、分离、纯化,制成纯度较高的提取物,然后采用提取物进行配液,制得注射液成品。配液过程环节较多,操作复杂,是影响痰热清注射液质量的关键过程,笔者实验室以近红外(near infrared, NIR)光谱为主要的技术工具,对这一过程所用的提取物质量及过程监控方法进行了系统研究[49],发现提取物质量和配液过程直接影响成品质量,需要进行更为深入的研究。

痰热清注射液成品的总含固量和灯检合格率是2个重要的质量指标,关系到产品的质量一致性和经济效益,但配液过程中影响这2个指标的关键影响因素却不够清晰,有待进行明确辨识,以便实施精确控制。Henriksen H C等[10]通过试验设计的方法分别研究了原料与过程变量对纸浆和奶酪产品质量的影响,采用近红外光谱的主成分得分值反映原料信息,结合蒸煮配方、蒸煮时间和切片质量等工艺参数,预测了纸浆的卡伯值;Jrgensen K等[11]则采用傅里叶变化中红外(FTIR)光谱的主成分得分值反映原料信息,结合蛋白质含量、凝乳时间、发酵剂添加,凝固物切割等工艺参数,对干酪粉(一种表征产量损失的参数)进行预测。类似方法对于中药产品质量参数的预测及其影响因素辨识具有一定的借鉴意义。但由于痰热清注射液配液过程操作步骤多、时间长、成本高,很难通过试验设计和实验室模拟的方法获取数据,而实际的批生产记录中则记录了较多的工艺参数数据,具有信息挖掘的加之。本文采集了24个批次的配液过程数据,包括5种提取物的NIR漫反射光谱和配液过程中的多项工艺参数,将各种提取物光谱的关键主成分得分值与各项工艺参数进行归一化处理后,建立回归模型,对痰热清注射液成品的总含固量和灯检合格率进行预测,初步辨识出了影响这2个质量参数的主要因素。, 百拇医药(李文龙 潘建超 薛东升 刘绍勇 瞿海斌)
