UPLCPDA同时测定藏药翼首草环烯醚萜苷和酚酸类10个化学成分的含量(1)

     [摘要] 建立超高效液相色谱法(UPLCPDA)同时测定藏药翼首草环烯醚萜和酚酸类10个化学成分的含量。采用Acquity UPLCR BEH C18(2.1 mm ×100 mm,1.7 μm)色谱柱,流动相0.2%磷酸水(A)乙腈(B),梯度洗脱,检测波长237,325 nm,流速0.4 mL·min-1,柱温30 ℃,进样量1 μL。研究结果表明UPLCPDA法定量分析翼首草10个化学成分在考察的范围内线性关系良好(r≥0.999 7);加样回收率为96.30%~103.0%(n=9),RSD为0.72%~2.9%(n=9)。所建立UPLC方法简便、准确、重复性好,可同时测定藏药翼首草中10个化学成分的含量。

    Simultaneous determination of ten major compounds including iridoid

    glycosides and phenolic acids in Pterocephalus hookeri by UPLCPDA

    [Abstract] This study is to develop an UPLCPDA method for determination of 10 major components in Pterocephalus. The UPLCPDA assay was performed on a Waters Acquity UPLCR BEH C18(2.1 mm ×100 mm,1.7 μm), and the column temperature was at 30 ℃. The mobile phase consists of water containing 0.2% phosphoric acid (A) and acetonitrile (B) in gradient elution at a flow rate of 0.4 mL·min-1. The detection wave length was set at 237 and 325 nm, and the injection volume was 1 μL in the UPLC system. The linear range of 10 detected compounds were good (r≥0.999 7), and the overall recoveries ranged from 96.30% to 103.0%, with the RSD ranging from 0.72% to 2.9%. The method was simple, accurate and reproducible, which can be used for the simultaneous determination of the content of ten major components in P. hookeri.

    翼首草(藏語名:榜孜毒乌)为川续断科匙叶翼首草Pterocephalus hookeri(C.B.Clarke)Heck 的干燥全草,收载于2015年版《中国药典》(一部)[1]及卫生部《藏药部颁标准》[2]中,为常用藏药。翼首草主要含有环烯醚萜类和皂苷类成分,如马钱苷、吴茱萸苷、齐墩果酸和熊果酸等[39],具有抗病毒、抗氧化及提高免疫力等作用。课题组前期对翼首草进行了LCQTOFMS的化学成分分析,首次发现翼首草中含有大量的马钱苷酸、异绿原酸A、异绿原酸C、林生续断苷Ⅰ、续断苷B和续断苷A,且这些化学成分具有较强的紫外吸收。因此,本文采用UPLCPDA对其环烯醚萜和酚酸类10个化学成分进行含量测定,为翼首草的质量控制提供新的科学依据。

    1 材料

    Waters Acquity UPLCTM超高效液相色谱仪,包括四元溶剂管理系统、样品管理系统、PDA检测器、Empower色谱工作站(美国Waters公司);BT125D 1/10万电子天平(德国Sartorius公司);KQ500E超声波清洗器(江苏省昆山市超声仪器有限公司)。

    马钱苷酸(111865201102)、绿原酸(110753201413)和马钱苷(111640201509)购自中国食品药品检定研究院;异绿原酸A(16031611)、异绿原酸C(16031613)购自成都曼思特生物科技有限公司;獐牙菜苷、林生续断苷Ⅰ、吴茱萸苷、续断苷B和续断苷A由本实验室分离制备;经光谱及高效液相色谱分析确认,纯度均在98%以上。HPLC级甲醇、乙腈(美国J.T.Baker公司),水为屈臣氏蒸馏水(可供UPLC/UPLCMS测定);HPLC级甲酸(美国Tedia公司)。甲醇、磷酸试剂均为分析纯。 (唐策文检王静 赵可惠 范刚孟宪丽 邹忠梅 张艺)
