[摘要] 采用超高效液相色谱-串联四极杆飞行时间质谱(UPLC-Q-TOF/MSE)结合诊断离子过滤方法对掌叶大黄中的酚类成分进行快速分析鉴定。首先,负离子模式下对酚类代表性单体没食子酸、(+)-儿茶素、(-)-表儿茶素、(-)-表儿茶素-3-O-没食子酸酯和原花青素B2进行分析,综合文献报道,总结质谱裂解途径,确定诊断离子;然后,应用诊断离子过滤快速筛选掌叶大黄提取液中的酚类成分,结合保留时间、质谱碎片信息、裂解行为和精确质量数(计算分子式)对化合物进行结构鉴定。在掌叶大黄中共鉴定了63个酚类成分(36个简单酚酸类化合物、8个类黄酮类化合物和19个鞣质类化合物),其中包括6个潜在的新化合物。诊断离子过滤方法可对大黄中酚类成分进行快速分析,并完善了中药大黄的药效物质基础。
