香青兰总黄酮对ApoE—/—小鼠动脉粥样硬化病变中TGFβ1/Smad信号通路及基质金属蛋白酶表达的影响(1)
[摘要]研究香青兰总黄酮(TFDM)对高脂饮食诱导载脂蛋白E基因敲除(ApoE-/-)小鼠动脉粥样硬化形成的影响及其相关机制。采用40只SPF级8周龄雄性ApoE-/-小鼠予高脂饲料喂养,随机分为5组:香青兰总黄酮低、中、高剂量组分别给予21,42,84 mg·kg-1·d-1灌胃;辛伐他汀组灌服辛伐他汀35 mg·kg-1·d-1;模型组灌服等剂量生理盐水。另设8只相同基因背景和周龄的雄性C57BL/6J小鼠为对照组予普通饲料喂养。各组均灌胃干预12周,采用HE染色观察主动脉病理形态学变化;qRTPCR法检测组织中TGFβ1,Smad2,Smad3,MMP2和MMP9基因的水平。实验结果显示:与模型组比较,HE染色观察TFDM给药组表现主动脉粥样硬化组织损伤明显减轻;TFDM不同剂量组可在一定程度上抑制高脂饲料诱导的ApoE-/-小鼠主动脉TGFβ1,Smad2,Smad3,MMP2,MMP9 mRNA的表达,且呈现一定的剂量依赖关系趋势。提示TFDM具有减缓小鼠动脉粥样硬化形成的作用,其作用机制可能与干预TGFβ1/Smad信号转导有关。

動脉粥样硬化(atherosclerosis,AS)是一种脂质介导的慢性血管炎性病变,是众多心、脑血管疾病发生和发展的重要病理基础,可对人类健康产生严重危害。TGFβ/Smad通路在AS的发生发展过程中发挥着重要的作用,TGFβ具有多种生物学的功能,其对动脉内皮的损伤,血管平滑肌的迁移,动脉壁脂质聚集,细胞外基质的沉积和炎性细胞的浸润等AS的关键步骤均有调节作用[13]。
香青兰Dracocephalum moldovica L,为唇形科青兰属植物,是维吾尔民族药之一,主要用于治疗气管炎、心脏病、高血压等疾患,具有补脑安神、活血化瘀、通路开窍、镇咳止喘等作用[4]。其主要化学成分有黄酮类、挥发油、萜类、氨基酸、微量元素及多肽等,香青兰总黄酮(D moldavuca total flavones,TFDM)为香青兰的主要有效成分,具有降血脂、抑制血小板聚集及保肝、抗炎等作用[57]。本研究应用高脂饲料喂养ApoE-/-小鼠建立AS动物模型,采用HE染色观察TFDM对主动脉斑块面积的干预作用,以及qRTPCR检测小鼠主动脉TGFβ1,Smad2,Smad3,MMP2和MMP9 mRNA的表达探讨TFDM的抗AS作用及其可能机制。
