四君子汤总多糖的提取条件优化与体外抗肿瘤作用研究

     【摘 要】目的:研究四君子汤总多糖提取的最佳工艺条件与体外抗肿瘤作用。方法:采用蒽酮-硫酸法测定总多糖的含量,以提取温度、提取时间、提取次数、溶剂用量为4个因素,分别设计3个水平进行正交实验;MTT法检测四君子汤总多糖对人肝癌细胞株HepG2的影响。结果:最佳工艺为15倍量水,95℃提取3小时,共4次;四君子汤总多糖在体外对HepG2细胞无明显的影响。结论:提取条件对于四君子多糖的得率具有影响;四君子汤总多糖没有直接的肿瘤抑制作用。

    【Abstract】Objective: To optimize the conditions of extraction for polysaccharide of Sijunzi decoction and to study anti-tumor activity in vitro. Methods: Total carbohydrate content was determined by anthrone-sulfuric acid method, the extraction temperature, extraction time, extraction times and solvent volume were designed by an orthogonal test method. The effects of Sijunzi decoction polysaccharides on human hepatoma carcinoma cell HepG2 was observed in vitro by MTT method. Results: It was optimal conditions that Sijunzi decoction was extracted with hot water (w/v of 1:15) at 95 ℃ every 3h for three times. The total polysaccharides from Sijunzi decoction had no obvious influence on HepG2 cells.Conclusion: The yield of polysaccharide was influenced by extraction conditions. Sijunzi decoction polysaccharides have no direct tumor-inhibiting activity in vitro.
