【摘要】目的 通过检测槟榔碱对胎鼠外周血细胞DNA损伤的程度和观察胎鼠生长发育情况，探讨槟榔碱对小鼠胚胎毒性的影响。方法 使用槟榔碱给雌鼠灌胃6—8周后，与雄鼠合笼，孕16d后各组随机取孕鼠6只，剖腹取胎鼠，检测各组胎鼠的外周血细胞DNA损伤情况，并进行分析。孕19d后处死各组孕鼠，检测胎鼠的生长发育情况以及各组胎鼠的畸形发生率。结果 正常的对照组胎鼠外周血细胞DNA出现典型的原型，而实验组的胎鼠外周血细胞DNA出现特征性的彗星形态。于孕19d处死各组动物，发现实验组动物胎鼠的生长发育指标包括身长、体重、尾长小于对照组，畸形发生率高于对照组。结论 槟榔碱能对胎鼠外周血DNA造成一定程度的损伤，并对胎鼠各项生长发育指标均有影响，推测槟榔碱对胚胎产生一定毒性影响。

The Effect of Arecoline on the DNA of Fetal Rat’s Peripheral Blood Cell

【Abstract】Objective To study the effects of arecoline on the embryo toxic by means of examine the damage of DNA in the peripheral blood cell andfetus’growth developmentMethods Keep giving mice arecoline for 6-8 weeks by oral administration, let female mice get pregnate. After 16d later,get the fetus from6pregnant rats of each group, examine and analysis the damage of DNA in the peripheral blood cell.after 19d, get all the rest of fetus, recording the growth,development and the rate of the foetal deformities. Results the DNA of contrast group’s form is normal, but the DNA of experimental group has been damaged. The growing development include the length, weight, tail length and the rate of foetal deformities are irregular than contrast group. Conclusion The arecoline have got definite damage action on DNA in Peripheral Blood Cell of fetus and have effects on fetus’ growing development, the result suggested that arecoline possessed some embryo toxic.

1 引言
全世界约有6亿槟榔嗜好者，而且近年来嚼食人数还有增加趋势。槟榔碱是从天然棕榈科植物槟榔的种子中提取的一种胺类生物碱，槟榔的化学成分复杂，而槟榔碱是槟榔的主要成分,有一定毒性。关于槟榔碱的毒理学作用已有许多报道，以往的研究表明，槟榔碱对小鼠骨髓细胞的DNA有一定的损伤作用, 具有一定的遗传毒性 ，对胚胎发育具有延迟作用。但对于槟榔碱对是否会对孕鼠产生一定毒性反应，是否会通过孕鼠影响到胎鼠的相关报道很少，而生殖毒性却是在如今越来越受到人们的关注。本文旨在探讨槟榔碱对小鼠妊娠毒性和胎鼠外周血细胞DNA的影响。
2 实验材料
2.1 实验小鼠
6—8周龄雌性昆明小鼠，购于湖南农业大学实验动物中心。
2.2 仪器
电泳仪： 北京君意东方电泳有限公司
荧光倒置显微镜：日本OLYMPUS公司
2.3 主要试剂
氢溴酸槟榔碱( arecoline hydrobromide ,AREC) ,购自Sigma 公司
彗星实验试剂盒：购于普洛麦格(北京)生物技术有限公司
SCGE碱性电泳缓冲液(pH13.0)：NaOH 12g，EDTA0.37224g，加ddH2O定容至1000ml ......
