雷米普利对2型糖尿病大鼠肾脏氧化应激的影响(1)

     [摘要] 目的 观察硝基酪氨酸在2型糖尿病模型大鼠肾脏的表达及雷米普利(Ramipril)对它的影响。方法 将大鼠分为正常对照组(N)、糖尿病组(D)、雷米普利治疗组(R),比较各组大鼠肾组织中丙二醛(MDA)含量、超氧化物岐化酶(SOD)活性,和硝基酪氨酸(NT)含量,同时观察血糖、血胰岛素、肾功能和肾脏形态变化。结果 与正常组比较,糖尿病组肾组织中MDA含量明显上升(P<0.05),SOD活性显著下降(P<0.05),NT增多(P<0.05),肾功能和肾脏形态受损,而雷米普利治疗组的上述表达较糖尿病组明显改善。结论 硝基酪氨酸在糖尿病肾病中的表达明显增加,而雷米普利可能通过抑制氧化应激反应对2型糖尿病模型大鼠肾脏产生保护作用。

    The Effects of Ramipril on Expression of Nitrotyrosine in the Kidney of Type 2 Diabetic Rats

    [Abstract] Objective To evaluate the expression of nitrotyrosine in the kidney type2 diabetic rats and observe the effect of Ramipril on nitrotyrosine. Methods 30 rats were divided into 3 groups of normal rats(N),diabetic rats(D),diabetic rats treated with Ramipril (R).Content of malonaldehyde(MDA),activity of antioxidant enzymes including superoxide edismutase(SOD) and nitrotyrosine(NT)in the renal tissue were compared among the groups. In addition, bloodglucose, serum creatinine(Cr)bloodureanitrogen(BUN)were measured; the renal tissue were observed by light micros copy and electron microscopy. Results Compared with normal group, the level of BUN, Cr, MDA and NT in diabetic rats was significantly increased, but the activities of SOD were decreased notably. In addition, the renal pathologic detriment was significantly higher than normal controls. But those were improved in the treated group obviously. Conclusion Oxidative stresses take an important role in the development of diabetic nephropathy. The expression of Nitrotyrosine in type2 diabetic rats was increased. And antioxidation may be the possible mechanism of the protective action of Ramipril.

    氧化应激是指机体在遭受各种有害刺激时,自由基的产生和抗氧化防御之间严重失衡,从而导致组织损伤。最近研究显示,氧化应激与糖尿病及其并发症的发生、发展密切相关,应用抗氧化治疗可逆转氧化应激对组织的损伤,从而阻止或延缓糖尿病及其并发症的发生、发展。雷米普利上市以来,主要作为控制血压、治疗心力衰竭及抗动脉粥样硬化药。近年来,大量资料及体内外试验表明,其也有很强的抗氧化作用,而硝基酪氨酸(nitrotyrosine,NT)是自由基引起蛋白质损伤产生,可以作为氧化应激的特异性标志物 ......
