Box—Behnken响应面法优选金樱子提取工艺(1)

     摘要:目的 筛选金樱子提取工艺。方法 以金樱子总皂苷、总酚酸含量为评价指标,乙醇浓度、提取时间、料液比、提取次数为考察因素,采用Box-Behnken响应面设计,Design-Expert8.0.5软件分析试验数据,建立多元二次回归方程,筛选金樱子提取工艺参数。结果 筛选出金樱子的最佳提取工艺参数为:乙醇浓度62.79%,料液比13.12∶1,提取时间137.93 min,提取次数1.85次,考虑到实际操作方便及工业大生产的实用性,将提取工艺参数修订为乙醇浓度60%,料液比13∶1,提取时间2 h,提取次数2次。结论 本研究筛选出的参数可为金樱子的提取工艺提供依据。

    Optimization of Extraction Process of Effective Constituents from Rosae Laevigatae Frustus by Box-Behnken Response Surface Method JIANG Ying-chun, DENG Chong, LI Jing-li (School of Pharmacy, Shaanxi University of Chinese Medicine, Xianyang 712046, China)

    Abstract:Objective To screen extraction process for Rosae Laevigatae Fructus. Methods With the content of total saponins and total phenolic acids from Rosae Laevigatae Fructus as the evaluation indexes, ethanol concentration, extraction time, solid-liquid ratio and extraction times as investigation factors, the test data were analyzed by Design-Expert8.0.5 software, and a multiple quadratic regression equation was set up to screen extraction process parameters of Rosae Laevigatae Fructus. Results The best extraction process parameters of Rosae Laevigatae Fructus were as follows:ethanol concentration was 62.79%;solid-liquid ratio was 13.12∶1;the extraction time was 137.93 min;extraction times were 1.85 times. Considering the convenience for actual operation and the practicability of industrial production, the extraction process parameters should be ethanol concentration of 60%, revised solid-liquid ratio of 13∶1, 2 hours’ extracting time, and 2 extraction times. Conclusion Final selected parameters provide the basis for the extraction process of Rosae Laevigatae Fructus.

    金樱子为蔷薇科植物金樱子Rosa Laevigata Mickx.的干燥成熟果实,其味酸、涩,性平,具有固精缩尿止带、涩肠止泻的功效[1]。金樱子可作为利尿剂、镇咳剂,也可用于皮肤肿瘤、烧烫伤、神经衰弱、高血压、神经性头痛、慢性肾炎等的治疗[2]。金樱子含有多糖、皂苷、三萜、黄酮类、有机酸等多种化合物,其中金樱子皂苷、酚酸具有广泛的生理活性,如溶血、抗癌、抗炎、抗菌、抗病毒、降低胆固醇等作用[3]。本试验以金樱子总皂苷、总酚酸的含量为评价指标,通讯作者:邓翀,E-mail:fmmudz217@126.com

    采用Box-Behnken响应面法,筛选金樱子乙醇回流提取工艺参数,为金樱子的提取工艺提供进一步参考。

    1 仪器与试药

    HH-2型数显恒温水浴锅(常州国华电器有限公司),SQ8200HD型超声波清洗器(上海冠特超声仪器有限公司),GB204型电子天平(瑞士梅特勒-托利多公司,d=0.1 mg),UV-1102紫外分光光度计(上海天美科学仪器有限公司)。

    金樱子药材购于西安万寿路药材市场,经本院王薇副教授鉴定为蔷薇科蔷薇属植物金樱子Rosa Laevigata Mickx.的干燥成熟果实。没食子酸对照品(中国食品药品检定研究院,批号110831-200302),齐墩果酸对照品(中国食品药品检定研究院,批号130742-201058),所有试剂均为分析纯,水为蒸馏水。, http://www.100md.com(江迎春等)
