Abstract: Objective To establish an HPLC method for content determination of astragalin and isorhamnetin-3-O-glucoside in Nelumbo nucifera Gaertn. Methods The chromatographic separation was performed on a Diamonsil C18 column (200 mm × 4.6 mm, 5 μm); the column temperature was maintained at 30 ℃; the mobile phase was acetonitrile-0.5% formic acid solution as gradient elution; the flow rate was 0.8 mL/min; the detection wavelength was at 347 nm; the injection volume was 10 μL. Results Under above chromatographic conditions, astragalin and isorhamnetin-3-O-glucoside with adjacent other peaks were among the good separation. Astragalin had a good linear relationship in the range of 0.1005–2.01 μg (r=0.999 6); the average recovery was 100.96% (RSD=2.68%). Isorhamnetin-3-O-glucoside had a good linear relationship in the range of 0.026–0.52 μg (r=0.999 3); the average recovery was 98.57% (RSD=2.34%). Conclusion The method is convenient and reliable, which can provide evidence for the quality control and resource development of Nelumbo nucifera Gaertn. as a related medicine., 百拇医药(裴香萍 李慧峰 杜晨晖 刘海霞 张淑蓉)
