复方地芬诺酯灌胃的家兔功能性便秘模型制作(1)
摘要：目的 探讨复方地芬诺酯复制功能性便秘家兔模型的可行性。方法 模型组采用复方地芬诺酯混悬液经口灌胃15 d，空白组予以相应量的蒸馏水灌胃。观察家兔首粒黑便时间、24 h粪便颗粒数以及24 h粪便质量，以判断模型的复制情况。结果 与空白对照组家兔比较，模型组家兔的首粒黑便排出时间明显延长，粪便重量减轻，粪便颗粒数减少，并表现出不同程度的精神状态的改变。结论 复方地芬诺酯灌胃可用于制作家兔功能性便秘模型，但在造模过程中要尽量避免各种不利因素的影响。

Experimental Research on the Functional Constipation Rabbit Model Duplicated by Compound Diphenoxylate Tablet Lavage

Abstract：Objective To explore the feasibility on the functional constipation rabbit model duplicated by compound diphenoxylate tablet.Methods Mixed suspension of compound diphenoxylate tablet was used to conduct gavage through the mouth for 15 days in the model group. At the same time ，the control group was given the correspounding amount of distilled water lavage.The discharging time of the first grain of black feces，fecal weight and grains in 24 hours were observed to judge the replication of the model.Results Compared with the control group，the discharging time of the first grain of black feces of the model group was prolonged obviously.The fecal weight and grains were reduced，and showed different degrees of mental status change.Conclusion The gavage of compound diphenoxylate tablet can be used to make functional constipation rabbit model.However，we should avoid all kinds of unfavorable factors during the experiment.

功能性便秘(functional constipation，FC)，是指無器质性病因，没有组织结构异常或代谢障碍，又排除肠易激综合征的慢性便秘，是功能性胃肠病最常见的类型之一，主要临床表现为排便次数减少、粪便干结、排便费力等。目前认为它的发病与结肠运动功能障碍、肛门直肠功能障碍、精神心理异常等因素有关[2]。近年来对功能性便秘的机制研究多采用大鼠或者小鼠作为模型动物，鲜有家兔的便秘模型制作。本研究参考了相关文献，制作复方地芬诺酯家兔功能性便秘模型，并对其中出现的问题和不利因素进行归纳和探讨，报道如下。
1资料与方法
1.1一般资料

1.1.1实验动物 4月龄的家兔20只，雌雄各半，体重2.5～3.5kg，购自成都达硕生物科技有限公司(合格证号：0014892)，到实验室后首先进行1 w的适应性喂养。按照随机数字表法将家兔分为空白组、模型组2个组，每组10只，分组后每组家兔雌雄各半，体重无显著性差异。
1.1.2 实验药品 复方地芬诺酯(成都第一制药有限公司，生产批号：1403024)[3]，为止泻药，每片含盐酸地芬诺酯2.5 mg，硫酸阿托品0.025 mg。阿拉伯树胶粉(Gum arabic powder)(天津市化工有限公司，生产批号：20140725)。100目粉状活性炭(成都瑞丽神火木炭有限公司，生产批号：20140712)。
1.1.3实验器材 烧杯、玻棒、导尿管(用于灌胃[12-13])、开口器、家兔固定器、酒精灯、粪尿分离器、注射器。
1.2实验方法
1.2.1药液及墨汁制备 药液：将复方地芬诺酯片研磨成粉与30℃蒸馏水以1：1的比例配置成复方地芬诺酯混悬液，灌胃前充分摇匀。墨汁：精密称取阿拉伯树胶100 g，加入800 ml蒸馏水煮沸至溶液透明；取粉状活性炭50 g，精密称定后加入上述溶液中煮沸3次(即文火煮沸后关火，待溶液冷却至室温后再次文火煮沸，如此操作3次)，待溶液冷却至室温后加蒸馏水定容至1000ml，放于0℃～5℃的冰箱保存，使用前摇匀[4]。, 百拇医药(王欣月 李思宇 罗芳丽 张微 师维 黄活石 李瑛)
