大鼠阴道上皮细胞体外培养研究进展(1)
摘要:外阴阴道假丝酵母菌病(VVC)是由假丝酵母菌引起的机会性真菌感染,约5%~10%的VVC患者可因病情反复发作成为复发性外阴阴道假丝酵母菌病(RVVC)患者。目前认为免疫异常是导致RVVC的主要因素,确切机制不明。大鼠阴道上皮细胞的体外培养为该病免疫机制的研究提供更多可能,其培养方法主要有组织块法和酶消化法,一般认为酶消化法优于组织块法。动情前期和动情期体外培养获得的大鼠阴道上皮细胞数比动情后期和动情间期多,但尚未形成一致的结论。细胞培养基成分也日渐完善,本文就大鼠阴道上皮细胞体外培养的研究进展做一综述。

Abstract:Vulvovaginal candidiasis(VVC)is an opportunistic fungal infection caused by Candida,about 5% to 10% of VVC patients may be recurrent as a result of recurrent vulvovaginal candidiasis(RVVC)patient.At present,the immune abnormalities are the main factors leading to RVVC,the exact mechanism is unknown.In vitro culture of rat vaginal epithelial cells provides more possibilities for the study of the immune mechanism of the disease. The culture method is mainly composed of tissue block method and enzyme digestion method.It is generally believed that the enzymatic digestion method is superior to the tissue block method.The number of vaginal epithelial cells in the prenatal and estrous stage was higher than that in the late stage and the estrous stage,but the conclusion was not yet reached.Cell culture medium components are increasingly perfect.In this paper,we reviewed the progress in the study of vaginal epithelial cells in vitro.

外阴阴道假丝酵母菌病(vulvovaginal candidiasis,VVC)是由假丝酵母菌感染引起的外阴阴道炎症,在阴道感染中居第2位,仅次于细菌性阴道炎[1]。据资料统计,70%~75%的女性一生中至少患VVC 1次,其中40%~50%患有VVC的女性会有复发感染,5%~10%的女性一年内发作4次或4次以上而成为复发性假丝酵母菌外阴阴道炎(recurrent vulvovaginal candidiasis,RVVC)[2]。目前认为阴道局部免疫功能异常是导致RVVC的主要因素,但是确切发病机制尚不清楚。大鼠阴道上皮细胞的体外培养为该病免疫机制的研究提供更多可能,其培养方法主要有组织块法和酶消化法,国内外许多学者对这两种方法均有研究,一般认为酶消化法优于组织块法[3-4],动情前期和动情期体外培养获得的大鼠阴道上皮细胞数比动情后期和动情间期多[4-5],但因研究报道极少,尚不能得出确切结论,何种培养基成分更适宜大鼠阴道上皮细胞体外培养也待进一步研究证实,本文就大鼠阴道上皮细胞体外培养的相关研究进展综述如下。

1 培养方法
1.1组织块培养法
组织块培养法是大鼠阴道上皮细胞的传统培养方法,其基本过程包括麻醉、解剖、培养材料的准备、接种、置培养箱中培养等步骤,李雅钗等用组织块法原代培养大鼠阴道上皮细胞,结果显示组织块法操作简单,技术容易掌握,成功率高,但细胞爬出慢,效率低,原代培养周期时间长,一般需要数周时间,由于未分离上皮层与固有层,在上皮细胞生长的同时,常伴有成纤维细胞的混杂生长,且其传代培养成功率低,故在大鼠阴道上皮细胞的原代培养中应用逐渐减少。
1.2酶消化法
酶消化法的基本过程包括麻醉、解剖、取材、酶消化、中和、清洗、重悬、加入培养基、置培养箱中培养等步骤,虽然存在较难掌握酶消化时間、对细胞损伤大、操作繁琐、费用高等缺点,但由于其培养周期较短,而且可针对上皮细胞选择相应特异性酶,短时间内获取大量纯度高的细胞,减少成纤维细胞的混杂,逐渐被广泛应用于上皮细胞的原代培养。, 百拇医药(卢伟 祁文瑾)
