丹参素和羟基红花黄色素A促血管新生及抗大鼠心肌梗死作用研究(1)
摘要:目的 探讨丹参素和羟基红花黄色素A促血管新生及抗大鼠心肌梗死的作用。方法 结扎冠状动脉左前降支法,建立大鼠心肌梗死模型,随机将造模成功的大鼠共75只进行分组,分别为假手术组、模型组、丹参素组羟基红花黄色素A组及联合组,各15只,于术后1~7 d尾静脉注射给药,1 次/d,比较五组大鼠心功能。结果 模型组大鼠左心室收缩压、左心室舒张末期压力、左心室压力最大变化速率与假手术组相比,差异明显,P<0.05;单用及配伍丹参素、羟基红花黄色素A组与假手术组和模型组左心室收缩压、左心室舒张末期压力、左心室压力最大变化速率比较,差异明显,P<0.05;而联合丹参素、羟基红花黄色素A与单用相比,左心室收缩压、左心室舒张末期压力、左心室压力最大变化速率差异不明显,P>0.05。结论 与丹参素和羟基红花黄色素A单用相比,丹参素和羟基红花黄色素A利于进一步改善大鼠心功能,研究价值较高。

Abstract:Objective To investigate the effects of danshensu and hydroxysafflor yellow A on promoting angiogenesis and anti-myocardial infarction in rats.Methods A method of reducing the left coronary artery ligation,establish a rat model of myocardial infarction,random model rats were a total of 75 groups were sham operation group, model group,Salvia miltiorrhiza group hydroxysafflor yellow A group and combined group,15 rats each.After intravenous injection of 1~7 d administration of 1/d,compared five groups of cardiac function in rats.Results The rats in the model group left ventricular systolic pressure,left ventricular end diastolic pressure,left ventricular pressure maximal rate compared with the sham operation group significantly,P<0.05;Single use and compatibility of tanshinol, hydroxysafflor yellow A group and sham operation group and model group left ventricular systolic pressure,left ventricular end diastolic pressure,left ventricular pressure,the maximum rate of change significantly,P<0.05;United tanshinol,hydroxysafflor yellow A and single phase ratio,left ventricular the systolic blood pressure,left ventricular end diastolic pressure,left ventricular pressure maximal rate was not significantly different,P>0.05.Conclusion Compared with Danshensu and hydroxy safflower yellow A alone,Danshensu and hydroxy safflower yellow A are beneficial to further improve cardiac function in rats,and have higher research value.

在心肌梗死治疗中,通过药物促进心脏自身搭桥属于常见的治疗手段,即利用药物促进缺血心肌部位的促血管生长因子表达,促进新血管生长,增强冠状动脉分支或侧支循环,具有改善患者心肌缺血缺氧作用[1]。中医学中认为,丹参和红花对“胸痹”、“真心痛”等指标有明显的效果,而丹参素和羟基红花黄色素A作为相应药物的重要活性成分,能够有效扩张血管,改善组织抗缺氧能力[2]。为了进一步了解丹参素和羟基红花黄色素A的价值,本次研究对丹参素和羟基红花黄色素A促血管新生及抗大鼠心肌梗死的作用进行了探讨,并选择75只大鼠作为研究资料,具体研究过程报道如下。
1 资料与方法
1.1 一般资料
选择清洁级健康SD大鼠75只作为研究资料,均为雄性,体重在220~260 g,平均體重(242±5)g。选择成都泰盟科技HX-100E小动物呼吸机、BL-420S系列生物机能实验系统,德国Leica仪器有限公司LMT260-XY图像采集系统,美国BIO-RAD公司凝胶成像系统、电泳仪、转膜仪;日本奥林巴斯IX71+DP72倒置显微镜,德国Eppendorf5804R冷冻离心机。丹参素(质量分数≥98%,生产企业:西安鸿生生物有限公司,批号:110516)、羟基红花黄色素A(质量分数≥98%,生产企业:成都贝斯特试剂有限公司,批号:BRS011748)。, 百拇医药(李江波)
