CDX2和CYP24A1在大肠癌中的表达(1)
摘 要:目的 通过研究CDX2、CYP24A1在大肠癌中的表达研究,探讨CDX2和CYP24A1的相关性。方法 应用免疫组化S-P法检测100例大肠癌组织及20例正常大肠癌组织中CDX2和CYP24A1的表达。结果 大肠癌组织中的CDX2和CYP24A1的蛋白表达低于正常大肠癌组织中的表达(P<0.05), 以正常大肠组织作为对照,CDX2 的蛋白水平表达量为100.00%, 癌旁组织和大肠癌组织的 CDX2 蛋白的相对表达量分别为77.12%、46.65%。CYP24A1 的蛋白水平表达量为 43.81%, 癌旁组织和大肠癌组织的 CYP24A1 蛋白的相对表达量分别为75.00%、96.82%。通过Pearson相关分析显示CDX2蛋白表达水平与CYP24A1蛋白表达水平之间呈显著负相关关系(P<0.05)。结论 CDX2和CYP24A1是具有较高敏感性的肠上皮特异性标志物,联合检测CDX2和CYP24A1的表达有助于鉴别良恶性大肠肿瘤。

Abstract:Objective To study the expression of CDX2 and CYP24A1 in colorectal cancer and to explore the correlation between CDX2 and CYP24A1.Methods The expression of CDX2 and CYP24A1 in 100 cases of colorectal carcinoma and 20 cases of normal colorectal carcinoma were detected by immunohistochemical S-P method.Results The expression is lower than that of normal colorectal carcinoma in colorectal carcinoma CDX2 and CYP24A1 protein expression(P<0.05),in normal colorectal tissues as the control group, the protein level of CDX2 expression was 100.00%, the relative expression of carcinoma and colorectal cancer CDX2 protein were 77.12% and 46.65%.The protein level of CYP24A1 was 43.81%,and the relative expression of CYP24A1 protein in para cancer tissue and colorectal carcinoma tissue was 75.00% and 96.82%,respectively.The Pearson correlation analysis showed a significant negative correlation between CDX2 protein expression and CYP24A1 protein expression(P<0.05).Conclusion CDX2 and CYP24A1 are highly specific intestinal epithelial specific markers.The combined detection of CDX2 and CYP24A1 expression is helpful to distinguish between benign and malignant colorectal tumors.

近些年,我国大肠癌发病率迅速上升,在黑龙江地区大肠癌发生率位居第二位。随着分子生物学,基因学的发展,人们逐渐认识到只有从基因水平着手才能从根本上治愈肿瘤[1] 。CDX2和CYP24A1分别是近年来国内外报道的与肠癌相关的分子标记物,但研究成果较少,还需进一步研究证实。CDX2 (caudal type ho-meobox transcriptionfactor 2)是一种新发现的特异性的核转录因子对肠道上皮细胞增殖、 分化的调控起着重要作用,可能具有抑癌基因的作用。 已有研究表明,CDX2 对大肠癌在内的消化道恶性肿瘤的侵袭和转移具有抑制作用,但具体机制尚未明确[2-4]。有研究报道活性维生素D(1α,25(OH)2D3)不仅能够抑制细胞增殖、诱导细胞分化与凋亡、增强免疫调节作用,还可以降低癌症发生风险[5,6],维生素D的限速酶CYP24A1是一个潜在的癌基因,在乳腺癌、肺癌、食管癌等组织中异常表达且与肿瘤恶性程度及患者不良预后相关[7-9]。本研究通过免疫组化法探讨CDX2与CYP24A1在大肠癌发生的过程中的作用及的相互关系,为临床诊断、治疗大肠肿瘤提供依据。, 百拇医药(王琳琳 刘蕾)
