温度与时间对尿液分析仪检测潜血与血尿测定的影响(1)
摘 要:目的 探究温度与时间对尿液分析仪检测潜血与血尿测定的影响,为提高检测的准确率寻求指导依据。方法 选取2016年3月~2017年3月我院门诊就诊患者留取血尿标本为研究对象,在不同室温下放置不同的时间后,用尿液分析仪和显微镜下红细胞计数,比较各组间隐血测试情况和红细胞的减少情况。结果 各组间隐血测试无改变,而镜检红细胞数随时间、温度的改变逐渐减少。相同来源的样品在12~22 ℃条件下检测结果优于22~30 ℃,差异有统计学意义(P<0.05)。结论 尿液标本在12~22 ℃室温条件下保存时建议在2 h内完成检测,若室外温度较高时,应当在1 h内完成检测。

The Effect of Temperature and Time on the Detection of Occult Blood and Hematuria by Urine Analyzer

Abstract:Objective To explore the influence of temperature and time on urine occult blood and hematuria determination by urine analyzer,so as to provide guidance for improving the accuracy of detection.Methods The hematuria was selected as the study object in the outpatient department of our hospital in March 2016~March 2017.After different room temperature at different room temperature,the urine analyzer and the erythrocyte count under microscope were used to compare the occult blood test and the reduction of red blood cells in each group.Results There was no change in the occult blood test among the groups,but the number of microscopic red blood cells changed gradually with time and temperature.The samples from the same source were better than 22~30°C at 12~22°C,and the difference was statistically significant(P<0.05).Conclusion When urine specimens are stored at room temperature 12~22°C,it is recommended to complete the test within 2 h.If the outdoor temperature is high,the test should be completed within 1 h.

血尿是臨床上多种疾病的共同病症,临床对于血尿的诊断主要是以红细胞和潜血这两种指标进行评判,但在大部分的实际操作过程中,由于仪器检测的敏感性较强,所以经检测的潜血和血尿镜检查的红细胞数量相差甚远,且不成正比[1]。其原因在于血尿检查过程中存放的时间、pH、温度和比重等都会受到影响。红细胞一旦受到破坏,镜检下观察到的红细胞数量就会减少,与潜血结果不一致[2]。我院通过对500例门诊患者的血尿镜检和潜血进行了检测和对比,以此证实尿液镜检即便是没有红细胞,潜血也不一定是阴性,现将结果作如下报道。
1资料与方法
1.1一般资料选取2016年3月~2017年3月昆明市官渡区人民医院门诊行血尿镜检和潜血镜检的500例患者为研究对象,其中男286例,女214例;年龄19~63岁,平均年龄(38.52±8.64)岁。使用仪器Mission U500尿液分析仪,与分析仪配套的尿液检测试纸,采用日本OLYMPUS公司生产的显微镜。对所有患者的尿液检查结果进行分析比较。
1.2方法取血尿样本后,即刻加入人血细胞计数板,并平放静止,置于高倍镜下观察计数5个高倍视野中红细胞的数量,并将试纸条即刻进行测试,观察是否出现隐血,再将尿液分成两份,标注1号与2号以作区别,将1号样品置于温度为12~22 ℃的环境中,2号样品置于温度为22~30 ℃环境中,每隔1 h将样品混匀后,置于井下和上机进行一次观测,并对结果进行登记分析。
1.2.1尿液分析仪检验 首先将新鲜混匀的10 ml尿液取出放入试管中,将试纸条直接浸泡在所选尿液中1 s,然后将试纸条置分析仪上进行检测,打印检验结果。, http://www.100md.com(朱春霖)
