三仁汤对脾胃湿热证型胃癌癌前病变模型大鼠的作用机理(1)
摘要:目的 探究三仁湯对脾胃湿热证型胃癌癌前病变模型大鼠关性的作用机理。方法 将50只健康清洁级SD大鼠视为研究对象,经数字随机法分为随机分为正常组、模型组、高剂量三仁汤组、中剂量三仁汤组和低剂量三仁汤组,总共5组,每组各10只,利用复合因素复制脾胃湿热证癌前病变模型大鼠。采用免疫组织化学检测法检测P53、Bcl-2;采用原位末端标记法检测细胞凋亡指数(AI);采用酶联免疫吸附法(ELISA)检测白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、白细胞介素8(IL-8)、白细胞介素10(IL-10)和环氧化酶2(COX-2),并进行组间比较和分析。结果 在P53、Bcl-2、P21ras、AI、COX-2水平方面,模型组要明显高于正常对照组(P<0.05);三仁汤(高、中、低剂量)组均明显低于模型组(P<0.05),以中剂量组降低效果最为明显。在血清IL-1β、IL-6、IL-8、IL-10水平方面,模型组要明显高于正常对照组(P<0.05),三仁汤(高、中、低剂量)组均低于模型组(P<0.05),以中剂量组降低效果最为明显。结论 三仁汤对脾胃湿热证型胃癌癌前病变模型大鼠关性的作用机理能表现为下调脾胃湿热模型大鼠胃黏膜P53、P21ras、Bcl-2、AI、COX-2蛋白水平的,抑制血清IL-1β、IL-6、IL-8、IL-10过度反应。

【Abstract】Objective: To explore the mechanism of Sanren Decoction on gastric precancerous lesions in rats with spleen-stomach damp-heat syndrome. Methods: 50 healthy and clean SD rats were regarded as the research objects and were randomly divided into normal group, model group, high-dose, medium-dose and low-dose Sanren Decoction groups. 5 groups in total, 10 rats each group. The model rats were made by using compound factors to replicate precancerous lesion with spleen-stomach damp-heat syndrome. The immunohistochemical detection method was used to detect P53 and Bcl-2 and the in situ end labeling method to detect cell apoptosis index (AI), enzyme-linked immunosorbent method (ELISA) to detect interleukin 1β (IL-1β), leukocyte mediator Interleukin 6 (IL-6), Interleukin 8 (IL-8), Interleukin 10 (IL-10) and Cyclooxygenase 2 (COX-2). The comparison and analysis were made among the groups. Results: In terms of levels of P53, Bcl-2, P21ras, AI, and COX-2, the model group was significantly higher than the normal control group (P<0.05) and Sanrentang groups (high, medium, and low dose) were significantly lower than the model group (P<0.05). The medium-dose group had the most obvious reduction effect. In terms of serum IL-1β, IL-6, IL-8 and IL-10 levels, the model group was significantly higher than the normal control group (P<0.05) and the Sanrentang groups (high, medium, and low dose) were all lower than the model group (P<0.05). The medium-dose group had the most obvious reduction effect. Conclusion: The mechanism of action of Sanren Decoction on gastric precancerous lesions in model rats with spleen-stomach damp-heat syndrome can be represented by down-regulating the gastric mucosal P53, P21ras, Bcl-2, AI, COX-2 protein levels in the model rats with the spleen and stomach damp-heat syndrome and inhibiting the overreaction of serum IL-1β, IL-6, IL-8 and IL-10., 百拇医药(邹先明 吴莹 吴欢)
