玉米苞叶中芦丁的含量测定(1)

     【摘 要】 目的: 建立HPLC法测定玉米苞叶中芦丁含量的方法。方法:采用高效液相色谱法,以甲醇为提取溶剂,超声辅助提取玉米苞叶中的芦丁,色谱柱为phenomenon C18 (5μm,250mm×4.6mm);检测波长为280nm;流动相采用乙腈-0.1%冰醋酸(20∶80);流速为0.8mL/min;柱温为25℃;进样量10μL。结果:芦丁进样量在0.19470~2.0047μg·mL-1范围内与峰面积呈良好的线性关系(r=0.9996),平均加样回收率为97.70%,RSD为1.46%(n=6)。结论:该测定方法简便、快捷,重现性好,可为玉米苞叶药材质量控制和评价提供参考。

    Determination of Rutin in Corn Bracts

    Abstract:Objective To establish a HPLC method for determination of rutin in corn husks. Methods High performance liquid chromatography (HPLC) was used to extract rutin from corn husks with methanol as the extraction solvent. The chromatographic column was phenomenon C18 (5μm,250mm×4.6mm),the detection wavelength is 280nm;the mobile phase was acetonitrile-0.1% glacial acetic acid (20∶80);the flow rate was 0.8mL/minccolumn temperature is 25 ℃;sample quantity is 10μL.Results There was a good linear relationship between the amount of rutin injected and the peak area within the range of 0.19470~2.0047μg·mL-1 (r=0.9996), and the average recovery rate was 97.70%. RSD was 1.46% (n=6).Conclusion the method is simple, rapid and reproducible, which can provide reference for quality control and evaluation of bract.
