不同加工方法对杜仲桃叶珊瑚苷含量的影响(1)

     【摘要】目的:比较不同加工方法对杜仲桃叶珊瑚苷含量的影响,为杜仲的产地加工提供科学依据。方法:采用高效液相色谱法测定桃叶珊瑚苷量,色谱条件:Diamonsil C18色谱柱(250 mm × 4.6mm,5 μm),流动相甲醇-水溶液(10∶[KG-*2]90),检测波长203nm,柱温25℃,流速1.0mL/min,进样量10μL。结果:不同加工方法杜仲桃叶珊瑚苷含量有差异。同一干燥方法,未发汗杜仲桃叶珊瑚苷量高于发汗杜仲;同一性状,烘干、晒干、阴干三者间桃叶珊瑚苷量具显著差异,以烘干最高,晒干次之,阴干最低;不同性状,板片状以100℃烘干、宽丝状以 60℃烘干含量最高。结论:不同加工方法对杜仲桃叶珊瑚苷含量影响显著,临床应用如以桃叶珊瑚苷的生物活性为主,结合实际生产,建议杜仲采用趁鲜切宽丝,不发汗,60℃烘干的产地加工方法。

    Abstract:Objective:To provide scientific basis for the processing method of Eucommia ulmoides Oliv by comparing the effect of different processing methods on the content of aucubin. Methods:Determine the content of aucubin in Eucommia ulmoides Oliv(EUO) by HPLC. A Diamonsil C18 column (4. 6 mm×250 mm,5 μm) was used for the analysis. The mobile phase was methanol-water(10∶[KG-*2]90),the flow rate was 1. 0 ml/min,the detection wavelength was at 203nm,and the column temperature was 25℃. Results:Eucommia ulmoides Oliv processed by different methods, the content of aucubin in them was various. Living on the same drying methods,the content of aucubin in herbs without undergoing the diaphoretic process was higher than the herbs experienced it. Herbs in the same trait,with different drying methods,there were significant difference in the content of aucubin,and the content in the drugs dried by oven was higher than the drugs dried by exposure to the sunor inthe shade. Herbs in different trait,the content of aucubin in the plate-like EUO samples dried in the oven in 80℃and the wile-filamentous-like EUO samples dried in the oven in 60℃was both higherthan all other like samples. Conclusion:There were significant difference in the content of aucubin of different samples processed by different methods. In the clinical application,if we considered the biological activity of aucubin as a focus,we suggested the EUO should be processed in wile-filamentous-like without undergoing diaphoretic process and dried in the oven in 60℃.

    杜仲为杜仲科植物Eucommia ulmoides Oliv.的干燥树皮,广泛分布于我国西南部和长江中上游地区,是我国的特产经济药材,具有补肝肾、强筋骨、安胎的功效,临床多用于肾虚腰痛、筋骨无力、妊娠漏血、胎动不安、高血压症等[1]。贵州产杜仲因其资源丰富,疗效显著,被业内公认为杜仲的地道产地,与天麻、灵芝组成的“贵州三宝”,是馈赠访友的必备之品,具有极高的药用和经济价值。产地加工作为控制药材品质的关键技术,杜仲虽有《本草经集注》“采皮阴干”、去粗皮晒干[1]和去粗皮“发汗”后晒干[2]的记载,但标准并不统一,质控指标单一。现有加工方法的研究多以降压活性成分的松脂醇二葡萄糖苷作为考察指标[3-4],对其他活性成分和药理作用的影响未见报道。

    杜仲的生物活性成分有木质素类、环烯醚萜类、黄酮类、多糖类及苯丙素类等,其所含桃叶珊瑚苷具有抗炎抗病毒、降血脂、抗氧化、抗疲劳、降压等广泛的药理作用[5-7],是杜仲的重要活性成分之一。研究以桃叶珊瑚苷为评价指标,考察不同性状、干燥方法及是否“发汗”对桃叶珊瑚苷含量的影响来探讨杜仲的产地加工方法,以期为全面评价和建立杜仲加工方法的质量标准体系提供科学依据。, http://www.100md.com(刘汇丽 魏学军 何鑫)
