积雪草甙对兔耳增生性瘢痕TGF-β1mRNA表达的影响(1)
[摘要]目的:通过建立兔耳腹侧增生性瘢痕的模型,探讨积雪草甙(Asiaticoside,Ad)对TGF-β1 mRNA表达的影响。方法:新西兰大白兔20只(喂养过程中死亡4只),在兔耳腹侧制备直径为1cm、深达软骨膜的圆形瘢痕93个,随机分为4组:空白对照组、得宝松组、低浓度积雪草甙组(25mg/ml)、高浓度积雪草甙组(50mg/ml),分别于瘢痕制备3天后局部应用相应药物,2周后取材,分别测量瘢痕组织TGF-β1 mRNA的表达量。结果:积雪草甙组与空白对照组相比,TGF-β1mRNA表达量明显降低,有明显统计学差异(P<0.01);积雪草甙与阳性对照组相比,二者mRNA表达量无统计学差异(P>0.05);积雪草甙组之间,高浓度组与低浓度组相比有统计学差异(P<0.05)。结论:较高浓度积雪草甙注射液局部注射可以减少增生性瘢痕TGF-β1 mRNA的表达。

Abstract:ObjectiveTo investigate the effects of Asiaticoside on TGF-β1 mRNA expression in hypertrophic scars.MethodsNinety-three full-thickness round wounds (1cm in diameter on ventral side of rabbits ears)on rabbits' ears of 20 rabbits(four died) were created. Then rabbits were randomized into 4 groups: negative group, diprospan group, asiaticoside(25mg/ml)group andasiaticoside(50mg/ml)group. The corresponding injectant were given three days after the scar model was formed lasting for 7days.Samples were collected 2 weeks later and expressions of TGF-β1 mRNA of different groups were detected.ResultsAsiaticoside group has a lower expression of TGF-β1 mRNA compared with negative group(P<0.01),while there are no respective diversity(P>0.05) between asiaticoside group and masculine group. As to the two asiaticoside groups, group of higher concentration has a lower expression of TGF-β1 mRNA and the difference is significant.ConclusionHigher concentration of asiaticoside injected could inhibittheexpressionofTGF-β1 mRNA

增生性瘢痕的形成是由于皮肤创伤后的愈合过程中成纤维细胞过量增殖和胶原纤维过度增生的结果,增生性瘢痕的防治是现代医学的难题之一,寻找治疗和预防瘢痕增生的药物具有重要的意义。积雪草甙为积雪草提取物三萜皂甙的主要成分之一,已有的资料显示,在各种皮肤损伤修复治疗中,无论是在正常创面,还是在延迟愈合的创面中,积雪草甙(Asiaticoside,Ad)均具有促进皮肤损伤修复的药物活性。临床上,已有积雪草甙的片剂和霜剂的应用,本实验是对兔耳增生性瘢痕的局部注射该提取物,通过RT-PCR方法对比研究其前后TGF-β1 mRNA的表达有无统计学差异,进而为临床上治疗增生性瘢痕寻找新的治疗途径。
1材料和方法
1.1 实验动物及主要试剂、仪器:新西兰大白兔20只,体重2.2~2.7kg,雌雄不拘,购自山东省实验动物中心,由青岛大学医学院附属医院实验动物中心代为饲养。适应性喂养1周,死亡2只,剩余18只。积雪草甙标准品,由西安市小草植物开发有限公司惠赠; SYBRPremixScript 逆转录试剂盒及定量PCR试剂盒(日本TaKaRa公司);RT-PCR引物: TGF-β1 mRNA 序列取自美国基因库,引物采用Primer5软件设计,所有引物由上海生工生物技术有限公司合成(见表2);冷冻高速离心机(日本Sigama公司)、7500 型荧光定量 PCR 仪(美国Bio-Rad公司)、普通离心机、研磨机、震荡仪、-70℃冰箱
1.2 兔耳瘢痕模型的制作:氯胺酮和地西泮按分别按2mg/kg和5mg/kg肌肉注射麻醉,然后在兔双侧耳腹侧分别作3~4个直径为1cm的圆形切口,切口间距1cm,完整切除皮肤全层至软骨膜表面,并切除软骨膜,保留耳软骨。创面压迫止血后予以暴露,无需包扎。术后3天死亡2只,剩余16只,共形成高出正常皮肤的瘢痕块93个。
随即选择每只兔的左右侧耳进行药物对比观察,分为空白对照组—低浓度积雪草甙组、空白对照组∣高浓度积雪草甙组、空白对照组∣得宝松组、低浓度积雪草甙组∣高浓度积雪草甙组、低浓度积雪草甙组∣得宝松组、高浓度积雪草甙组∣得宝松组;共得到空白对照组19个瘢痕块、得宝松组22个、低浓度积雪草甙注射组24个(25mg/ml)、高浓度积雪草甙注射组28个(50mg/ml),各组分别于瘢痕制作后3天开始隔天局部注射得宝松和相应浓度积雪草甙药液0.5ml/个,各组局部药物注射均持续至瘢痕制作术后10天。于瘢痕制作术后2周切取瘢痕组织标本。
1.3 HE染色:于瘢痕制作2周后切取瘢痕组织,随即固定于中性甲醛液,常规石蜡包埋,4μm切片,切片常规用二甲苯脱蜡,经各级乙醇至水洗,然后苏木素染色5min、自来水冲洗,盐酸盐酸乙醇化30s,置伊红液2min,再经常规脱水、透明、封片,镜检。, http://www.100md.com(赵文鲁 匡瑞霞 刘 肃 陈 璐)
