壳聚糖-胶原-CNTF复合神经导管修复大鼠周围神经缺损(1)

     【摘要】目的:探究由壳聚糖、胶原和睫状神经营养因子(CNTF)制成的复合神经导管修复大鼠周围神经缺损的可行性。方法:采用冷冻干燥法制备复合神经导管,并进行导管降解实验。取30只SD大鼠随机均分为3组:自体神经移植组(A组)、壳聚糖-胶原-CNTF复合神经导管组(B组)、壳聚糖-胶原导管加管内一次性给CNTF组 (C组) ,制作坐骨神经10 mm缺损,按上述分组进行桥接。术后3个月进行大体观察、神经电生理测定、组织学检测、图像分析。结果:壳聚糖-胶原-CNTF复合神经导管约13周完全降解。术后3个月时3组大鼠术侧后肢运动功能所改善,A组、B组较好,均好于C组。两组复合导管基本降解管内的再生神经己经完整地连接神经断端。电生理测定、组织学检测、图像分析显示:A组和B组神经传导速度、再生神经的数量、质量无显著性差异(P>0.05),而A组和B组数据均优于C组(P<0.05)。结论:壳聚糖-胶原-CNTF复合神经导管能有效的修复大鼠坐骨神经10 mm缺损,效果接近自体神经移植,优于管内一次性给CNTF的壳聚糖-胶原复合神经导管。

    chitosan-collagen-CNTF composite never conduit prepairing peripheral nerve defect in rats

    【Abstract】Objective:To explore the feasibility of chitosan-collagen-CNTF composite never conduit prepairing sciatic nerve defect in rats. Methods:Using freeze-dried method to make composite never conduits.the degradation test was perfoemed by weight loss measurements in 0.01mol/L phosphate-buffered saline (PBS) solution.Thirty Sprague-Dawley(SD) rats randomly were averagely divided into three groups, autograft nerve (group A), chitosan-collagen-CNTF composite nerve conduit (group B), chitosan-collagen composite nerve conduit and only once injection of CNTF into conduit (group C). Each rat underwent left sciatic nerve transection and left a 10 mm nerve defect, and then they were treated respectively according to the above mentioned three experimental groups. three months later after operation ,foot ulceration, gait, joint movement of rats were observed. composite never conduits and nerve regeneration were observed by Electrophysiological observation: nerve conductive velocity (NCV) was measured. Part of specimens were examined to know status about myelin sheaths by electron microscope.Results:The chitosan-collage-CNTF composite never conduits were fully degraded after about 13 weeks in PBS solution.
