姜黄素对结肠癌HCT116细胞侵袭和抗失巢凋亡的影响(1)
摘 要:目的 探讨姜黄素对结肠侵袭和失巢凋亡的影响。
方 法:应用姜黄素处理人结肠癌细胞系HCT116后,采用软琼脂集落培养试验检测锚着不依赖性增殖,采用Boyden小室模型试验检测癌细胞的侵袭性,采用琼脂糖凝胶电泳和TUNEL检测癌细胞失巢凋亡。
结 果:姜黄素可有效抑制结肠腺癌细胞集落生长和侵袭能力,且与浓度相关。琼脂糖凝胶电泳和TUNEL结果显示,姜黄素可诱导结肠癌细胞失巢凋亡,且与浓度和时间相关。
结 论:姜黄素可抑制人结肠癌细胞侵袭,诱导失巢凋亡是其机制之一。

Effects of Curcumin on Invasion and Anoikis Resistance of Human Colon Cancer Cell HCT116
ABSTRACT:AIM To study the effects of curcumin on anoikis resistance in human colon cancer cells.
METHODS: After colon cancer HCT116 cells were treated with curcumin,the growth of anchorage independence of colon cancer HCT116 cells was examined in soft agar colony formation,and apoptosis of colon cancer were evaluated by terminal deoxynucleatidyl transferase-mediated deoxyuridine triphosphate nick end labeling(TUNEL) and DNA ladder.
RESULTS:Curcumin could significantly inhibit the growth of anchorage independence of colon cancer HCT116 cells in soft agar colon foamation in a dose-dependent.Curcumin could induce apoptosis of colon cancer cells from DNA ladder and TUNEL assay,which were in a time- and dose- dependent manner.
CONCLUSIONS: Curcumin could inhibit the growth of anchorage independence of colon cancer HCT116 cells,which was related to anoikis.

细胞与细胞外基质(extracellular matrix,ECM)脱离黏附接触或者是接触不完全后出现的凋亡现象,被称为失巢凋亡(Anoikis)。“Anoikis”为希腊语“无家可归者(homelessness)”[1~3]。正常上皮细胞或内皮细胞脱离于细胞ECM的联系时会发生失巢凋亡,而大多数来源于上皮或内皮的肿瘤细胞则丧失了这种特性。恶性肿瘤细胞之所以有别于正常细胞,出现局部侵袭和转移,其重要原因之一是肿瘤细胞可以逃避失巢凋亡,即抗失巢凋亡。姜黄素(curcumin)系从中药姜黄中提取的酚性色素,有抗氧化、抗促癌、抗突变、抗炎、降血脂等广泛药理作用。近年来的研究表明,姜黄素可以预防和治疗多种恶性肿瘤[4-6]。但是尚不清楚姜黄素对结肠癌细胞失巢凋亡的影响。为此,我们采用姜黄素处理结肠癌HCT116细胞,探讨了其对抗失巢凋亡的影响,旨在了解姜黄素对结肠癌细胞的作用及可能的分子机制。
材料和方法
1.材料姜黄素(Curcumin),购自Sigma公司。以少量DMSO 融解,配置成10 mmol/L的溶液,置于-4℃保存。临用时,以完全细胞培养液稀释姜黄素到所需浓度,使DMSO终浓度
