银杏平颤方及其拆方对PD小鼠行为学形态学的影响(1)
摘 要:目的:探讨中药治疗帕金森病(PD)的作用和机制,观察银杏平颤方及其拆方对帕金森病(PD)模型小鼠行为学和形态学的影响。方法:应用MPTP制作PD小鼠模型。应用爬竿试验测试行为学变化和HE染色观察黑质和纹状体形态结构的变化。结果:(1)模型组行为学评分明显高于正常组(P<0.001);治疗14天、28天所有中药组行为学得分明显降低(P<0.001)。(2)HE染色显示脑组织形态结构改变也较明显,与行为学变化相应,但未进行统计学分析。结论:银杏平颤方及其拆方组能改善PD模型小鼠的行为学表现和影响黑质、纹状体的形态结构变化。

Abstract:Objective:To investigate the effect and its mechanism of Chinese herb recipe in the treatment of PD,to measure the effect of Ginkgo Biloba Pingchan Recipe and its three similar prescriptions on the behavioral test and neurons in substantia nigra compacta(SNc)and striatum of model mouse for PD.Methods:The Parkinson’s disease model was established by consecutive administration of MPTP to C57/BL mice. The doses of Ginkgo Biloba Pingchan Recipe were respectively given for 2 weeks and 4 weeks in the treatment groups. Animals were examined behaviorally with the pole test consecutively. then killed,Taking out the forebrain and striate body were observed in morphology by HE.Results:(1)Ginkgo Biloba Pingchan Recipe(GBPR) and its three similar prescriptions improved the dyskinesia of PD mice and showed different disability in pole the test and decrease of spontaneous motor activity, the model group mice demonstrated dyskinesia, scores obtained from the behavioral test were increased significantly compared with Normal control group. While the scores in treatment groups showed a marked decrease to some degree (allP<0.05). But there was no difference among the Chinese medicine groups(P>0.05).(2)GBPR and its three similar prescriptions protected neurons in SNc. The HE showed that, there were seldom neurons in SNc in model group;while there were much more neurons presented in the treatment groups than that in model group.Conclusion:Ginkgo Biloba Pingchan Recipe plays role in improving disability in pole test and increased spontaneous motor activity and in protection the neurons.

帕金森病又称震颤麻痹,是一种好发于中老年人[1]的多巴胺能神经元功能降低引起的锥体外系慢性病,现在多数学者认为与氧化应激-自由基有关[2-3]。原发性帕金森病的主要临床症状和体征为静止性震颤、肌强直、少动和体位障碍。有时可出现面目呆滞、发音低、书写字体过小、唾液增多、皮脂外溢、体温增高、出汗增多、便秘、疲乏、手脚痉挛引起的疼痛、抑郁及情感方面改变等。延缓PD功能障碍的抗氧化治疗有诱发精神障碍的副作用[4-5]。为了研究探讨PD的发病因病机,建立了不同的动物模型。以往的研究已证实MPTP小鼠模型的生物化学和组织化学特征均与临床PD相似。因此,本实验采用以MPTP腹腔注射诱发PD小鼠模型,观察PD小鼠行为学、病理学的变化,研究中药银杏平颤方及其拆方对PD小鼠行为及DA能神经元形态结构的影响,以探讨其防治PD的机制。
1材料与方法
