益气温阳护卫法对哮喘大鼠GATA3_mRNA表达的影响(1)
摘 要:目的:探讨益气温阳护卫法对哮喘大鼠Th细胞异常分化相关分子基因表达影响。方法:50只SPF级雄性SD大鼠随机分为正常对照组、哮喘模型组、益气温阳护卫组、地塞米松组和卡介菌多糖核酸组,每组10只。采用TaqMan Real time PCR测定各组外周血单个核细胞(PBMC)中GATA3_mRNA的表达变化。结果:哮喘模型组PBMC中GATA3_mRNA表达显著高于正常对照组(P<0.01),益气温阳护卫组、地塞米松组和卡介菌多糖核酸组PBMC中GATA3_mRNA的表达显著低于哮喘模型组(P<0.01),且与正常对照组PBMC中GATA3_mRNA的表达相当。结论:益气温阳护卫法调节Th1/ Th2细胞亚群趋于平衡可能是通过抑制哮喘大鼠GATA3_mRNA 的表达实现,从而达到防治哮喘的作用。

Effect of Yiqi Wenyang Huwei Method on Expression of GATA3 in Rat Asthma Model

Abstract:Objective:Explore the effect of Yiqi Wenyang Huwei(YWH) method on gene expression of Th cell differentiation-related abnormal in a rat asthma model.Methods: Fifty SPF male SD rats were randomly divided into 5 groups: the normal control group,the asthma group, the YWH group,the dexamethasone group,the BCG-PSN group.The mRNA expression of GATA3 in the PBMCof all groups were detected by TaqMan Real time PCR.Results: The mRNA expression of GATA3 in PBMC of asthma group were significantly higher than that of control group(P<0.01), the mRNA expression of GATA3 in PBMC of YWH group,the dexamethasone group and the BCG-PSN group was significantly lower than that of asthma group(P<0.01),but corresponding with the normal control group.Conclusion: YWH method regulating Th1/Th2 cell subsets may be balanced by inhibiting the mRNA expression of asthmatic rats GATA3 achieve, so as to achieve the role of prevention and treatment of asthma.

支气管哮喘是一种常见病、多发病,由多种炎性细胞参与的气道慢性炎症。现已明确,支气管哮喘是一种免疫缺陷导致的变态反应性疾病,表现为Th1和Th2细胞亚群失衡,即Th2型优势应答,这是支气管哮喘病理过程形成的始动因素和维持因素[1 ]。鉴于Th1、Th2是由共同的前体细胞(Th0)分化而来,而转录因子GATA3是特异性调控Th0分化、起着Th1/ Th2转换开关作用的关键因子, GATA3是Th2特异的转录因子, GATA3不仅在Th2细胞极化过程中起关键作用,而且它还是所有Th2型细胞因子转录所必需的[2]。研究表明,支气管哮喘气道中GATA3 表达显著增加,且这种表达增加与IL-5 呈正相关,FEV1 及AHR 呈负相关[3]。益气温阳护卫法由国家著名中医药专家洪广祥教授提出,是临床上防治支气管哮喘的有效方法。依据该法而拟定的益气温阳护卫汤由玉屏风散、桂枝汤及仙茅、淫羊藿组成。以往的研究已证实该方具有降低哮喘豚鼠及哮喘患者气道反应及明显延长哮喘发作的潜伏期的作用。降低哮喘豚鼠支气管肺泡灌洗液(BALF)嗜酸性细胞阳离子蛋白(ECP)的水平及减轻气道炎症,且明显优于玉屏风散组[4-7]。
本研究运用益气温阳护卫法干预哮喘大鼠,并设立对照,检测干预前后GATA3-mRNA表达的变化,拟从基因转录水平探讨该法调节Th1/Th2平衡的作用,进而探讨其治疗哮喘的机制,为科学指导用药提供依据。
1 材料与方法
1.1 实验动物 SPF级雄性SD大鼠40只,4~5周龄,体重120~170g,购自南方医科大学实验动物中心。按照随机原则分为5组,每组10只,自由进水与饮食,常规饲养1周后建立哮喘模型。动物合格证号:SCXK(粤)2006-0015。
1.2 实验用药 (1)实验用益气温阳护卫汤的制备:益气温阳护卫汤药材购自江西中医学院附属医院。将生黄芪、白术、防风、桂枝、白芍、生姜、大枣、炙甘草、仙茅、淫羊藿等10味药按比例2∶1.5∶1.5∶1∶1∶1∶1∶0.5∶1∶1.5配伍,取上述生药1200g。然后取8倍量冷水浸药0.5h,第1煎1h(沸后);第2煎加6倍量水煎40min,将两次煎液过滤合并浓缩,将提取物混合成合剂(其中白术、防风、桂枝、白芍、生姜提取挥发油),调至含生药2g/mL,浓度溶液600mL密封于无菌量瓶,存放于4℃冰箱。(2)地塞米松片,0.75mg/片,仙居制药股份有限公司。(3)斯奇康(卡介菌多糖核酸)注射液,湖南九芝堂制药有限公司。(4)用药剂量依据2000年版《中华人民共和国药典》,根据人和动物体表面积折算的等效剂量比率表计算,用药量相当于临床等效剂量。, 百拇医药(余建玮 薛汉荣 洪广祥 邱小华 邹叶青 程光宇 付向春 杨玉)
