活血健脾补肾法对MSC移植在结肠炎大鼠肠道增殖分化的影响(1)

    摘 要:目的:探讨中医活血健脾补肾法对骨髓间充质干细胞(MSC)移植在结肠炎大鼠肠道增殖分化的影响。方法:雌性SD大鼠以5%2、4、6,三硝基苯磺酸(TNBs)诱导形成结肠炎模型,造模后随机分为模型对照组、MSC移植组、活血组、健脾组、补肾组,第2天将体外分离培养的雄性SD大鼠MSC经尾静脉注射到各组雌性大鼠体内。疗程结束后,取大鼠结肠组织,免疫组化法检测其肠道干细胞标记物Musashi-1、端粒酶逆转录酶(TERT)的表达。结果:免疫组化检测到MSC移植组及中药治疗各组大鼠结肠黏膜组织Musashi-1及TERT均有阳性表达,其中活血组与MSC移植组比较差异有显著性意义。结论: 活血法代表方药桃红四物汤较明显促进MSC在结肠炎大鼠肠道增殖分化。

    Kidney Recipe on Proliferation and Differentiation of MSC in the Colon of

    Ulcerative Colitis After Transplantation

    Abstract:Objective:To explore the Effect of promoting blood flow,invigorating spleen or invigorating kidney recipe on proliferation and differentiation of Bone marrow mesenchymal stem cell (MSC) in the Colon of Ulcerative Colitis(UC) after transplantation. Methods:SD female rats were randomly into the model group,the MSC transplantation group,promoting blood fiow group,invigorating the spleen group and invigorating the kidney group after the colitis model were induced by Trinitrobenzene sulphonic acid,(TNBs).The MSC from SD male rats were purified by culture expansion in vitro and then were transplanted into SD female rat madels of UC through caudal vein next day.The expression of Musashi-1、TERT in colon tissue after the treatment was examined by immunohistochemistry.Results:The expression of Gastrointestinal stem cells markers such as Musashi-1 and TERT was positive, promoting blood flow group is significantly increased(P<0.05).
