定痫丸对戊四唑点燃癫痫大鼠脑内神经递质含量及海马c-fos表达的影响(1)

摘 要:目的:通过观察定痫丸对癫痫大鼠神经递质谷氨酸(Glu)、γ-氨基丁酸(GABA)和海马c-fos表达的影响来探讨该方治疗癫痫的机理。方法:采用腹腔注射戊四唑3.2mg/100g,连续给予4周造成大鼠癫痫模型,造模同时给予定痫丸、丙戊酸钠和定痫丸加丙戊酸钠治疗,连续4周。结果:采用戊四唑造模后,模型组大鼠脑组织中谷氨酸含量升高而γ-氨基丁酸含量明显降低,c-fos阳性细胞数明显增多,与正常组比较有显著性差异(P<0.05)。定痫丸组、丙戊酸钠组和中西药合用组均可明显降低谷氨酸含量,升高γ-氨基丁酸含量,减少c-fos阳性细胞数,与模型组比较具有显著性差异(P<0.05)。结论:定痫丸可对抗戊四唑致大鼠癫痫的作用,作用机理与降低脑内神经递质谷氨酸含量、升高γ-氨基丁酸含量以及阻断脑内c-fos蛋白的表达有关。

Abstract:Objective:Observe effect of Dingxianwan on quantity of GABA,Glu and c-fos in hippocampus so that we probe into the efficacy. Methods:Intraperitoneal injection of pentylenetetrazol 3.2mg/100g, consecutive to cause epilepsy in 4 weeks, At the same time to be cure by Dingxianwan,VPA, Dingxianwan combined with VPA. Results:After modeling by PIZ,model group were increased in brain tissue Glu, GABA concentration decreased, c-fos positive cells increased significantly. Dingxianwan group, VPA group and the Chinese and western medicine group were decreased Glu content, increased GABA levels, reduce the number of c-fos positive cells, which was indistinctive compared with model group. Conclusion:Dingxianwan can resist obviously seizure from PTZ-kindlod rats,its function mechanism may be that react by reducing the content of Glu and expression of c-fos in the hippocampi and increasing the content of GABA of PTZ-kindled rats.
