益气化瘀方对大鼠前列腺增生组织(1)

    摘 要:目的:观察益气化瘀方对大鼠前列腺增生组织血管新生和基质金属蛋白酶2(MMP-2)及其抑制物(TIMP-2)表达的影响。方法:50只SD大鼠去势7天后,皮下注射丙酸睾酮 5mg·kg-1·d-1,同时给予益气化瘀方、非那雄胺混悬液灌胃,30天后股动脉放血处死大鼠,摘取前列腺,测量前列腺体积,计算前列腺指数(PI),检测前列腺微血管密度(MVD)和前列腺组织中MMP-2及其抑制物(TIMP-2)表达。结果:与正常组比较,模型组大鼠前列腺指数明显增大(P<0.01),前列腺组织MVD明显增高(P<0.01),MMP-2蛋白表达显著增高(P<0.01),MMP-2/ TIMP-2比值明显增大(P<0.05);各给药组前列腺前列腺指数、MVD、MMP-2表达、MMP-2/ TIMP-2比值均明显低于模型组(P<0.05或P<0.01);益气化瘀方中、高剂量组大鼠前列腺指数、MMP-2表达量、MMP-2/ TIMP-2比值与非那雄胺组比较,差异均无统计学意义(P>0.05),但前列腺组织MVD明显低于非那雄胺组(P<0.05)。结论:中药益气化瘀方具有抗前列腺组织增生作用,其机制可能与抑制MMP-2过度表达,进而抑制前列腺增生组织新生血管形成有关。

    Effects of Yiqi Huayu Chinese Medicine on the Expression of MMP-2/TIMP-2 in BPH Rats

    Abstract:Objective:To study the effects of Yiqi Huayu chinese medicine on renew of new vessels and MMP-2 and TIMP-2.expression in BPH rats. Methods:Castrated 7 days later, Fifty SD rats were injected with testosterone(5mg·kg-1·d-1) ,the decotion of Yiqi Huayu and finasteride suspension were orally administrated respectively, after 30 days' treatment, the rats were sacrificed, the prostate volume were measured, and the prostate index(PI) were analyzed, the microvessels density(MVD) and level of MMP-2 and TIMP-2 were measured. Results:The levels of PI, MVD, MMP-2, and the ratio of MMP-2 /TIMP-2 of model group were obviously higher than the control group(P<0.01),but these The levels in all drug treatment groups were remarkably lower than the model group(P<0.05 or P<0.01).and There were no difference between the middle, high doses of Yiqi Huayu chinese medicine group and the finasteride group(P>0.05),but the level of MVD were remarkably lower as compared with the finasteride group(P<0.05). Conclusion: Yiqi Huayu could inhibits BPH, mechanisms underlying it may be related to restrain the levels of MMP-2, and renew of blood vessel. ......
