茶多酚预处理对β-淀粉样多肽致大鼠学习记忆障碍的改善作用(1)

    摘 要:目的:探索不同剂量茶多酚预处理对β-淀粉样多肽(Aβ)所致大鼠学习记忆障碍的影响。方法:将50只健康SD大鼠随机分为假手术组;模型组;茶多酚组(低、中、高3个剂量组)。采用Aβ25-35建立AD模型,给药组造模前2周按低、中、高3种不同剂量灌服茶多酚,造模后再连续灌服2周,每天1次。干预结束后,利用Morris水迷宫实验观察大鼠记忆能力改变。之后处死大鼠,检测各组海马部位Ach、ChAT及AchE的含量或活性。结果:模型组与假手术组相比,大鼠找到平台的时间及穿越站台所在位置的次数明显减少,Ach含量和ChAT活性明显降低,AchE活性有增加趋势,但与假手术组相比,没有显著差异(P>0. 05);给予不同剂量的茶多酚能不同程度改善AD大鼠的学习记忆障碍,提高AD大鼠Ach含量和ChAT的活性,与模型组比较有显著性差异,但对AchE活性影响不明显。结论:茶多酚能够改善AD模型大鼠学习记忆能力,提高大鼠脑组织ChAT活性,提示药物的作用可能与胆碱递质合成增加有关。

    Effect of Tea Polyphenols on Cognitive Impairment of Rats Model

    Induced by β-amyloid Peptide

    Abstract:Objective:To observe the effect and significance of different doses of tea polyphenols pretreatment on spatial memory and learning ability of Alzheimer rat model induced by β-amyloid peptide.Methods:Fifty rats were randomly divided into sham operation, model and model+tea polyphenols Low, medium and high dose group. The AD model rats were established by induced Aβ25-35; the treatment groups were adminstrated proper doses of tea polyphenols before operation and continued, once a day for 2 weeks. The changes of learning and memory ability of rats were observed by the morris water maze and shuttle box test. After the rats were killed, the contents or activity of Ach, ChAT and AchE in hippocampal area were detected. Results: Compared with sham operation group, the model group rats had extended escape latency, reduced crossing platform times(P<0.05).Compared with model group, three treatment groups shortened the escape latency and improved crossing platform times(P<0.05). Ach content and ChAT activity was significantly decreased, AchE activity increased, but with the sham group, no significant differences (P>0.05). Different doses of TP improved the learning and memory of AD rats , increased Ach content and ChAT activity , with the model group were significantly different (P <0.01), but the AchE activity was not obvious. Conclusion:Tea polyphenols can improve the ability of AD rats of learning and memory , enhance the ChAT activity of rat brain tissue, suggesting the drug may be related to the increased synthesis of the neurotransmitter acetylcholine. ......
