黄芩提取物工艺研究(3)

Ⅰ组:加浓HCl 3.7mL,调pH1~2,80℃水浴,恒温30min,沉淀,加350mL蒸馏水,搅匀(清),加4.4mL 40% NaOH,调至pH7,加350mL 95%乙醇,溶解,离心(4000转,20min)后,加浓HCl 7.0mL,调pH1.0~2.0,60℃水浴保温30min,静置,离心,沉淀,用700mL蒸馏水洗两次,水洗第二遍时pH5~6,700mL 85%乙醇洗两次,离心,沉淀,60℃真空干燥,得黄芩提取物。
Ⅱ组:加浓HCl 4.4mL,调pH1~2,80℃水浴,恒温30min,沉淀,加350mL蒸馏水,搅匀(清),加4.2mL 40% NaOH,调至pH7,加350mL 95%乙醇,溶解,离心(4000转,20min)后,加浓HCl 7.2mL,调pH1.0~2.0,60℃水浴保温30min,静置,离心,沉淀,用700mL蒸馏水洗两次,水洗第二遍时pH5~6,700mL 85%乙醇洗两次,离心,沉淀,60℃真空干燥,得黄芩提取物。
Ⅲ组:加浓HCl 5.2mL,调pH1~2,80℃水浴,恒温30min,沉淀,加120mL蒸馏水,搅匀(有黏液),加5.8mL 40% NaOH,调至pH7,加120mL 95%乙醇,溶解,离心(4000转,20min)后,加浓HCl 7.6mL,调pH1.0~2.0,60℃水浴保温30min,静置,离心,沉淀,用240mL蒸馏水洗两次,水洗第二遍时pH5~6,240mL 85%乙醇洗两次,离心,沉淀,60℃真空干燥,得黄芩提取物。
Ⅳ组:加浓HCl 5.2mL,调pH1~2,80℃水浴,恒温30min,沉淀,加120mL蒸馏水,搅匀(有黏液),加5.8mL 40% NaOH,调至pH7,加120mL 95%乙醇,溶解,离心(4000转,20min)后,加浓HCl 7.6mL,调pH1.0~2.0,60℃水浴保温30min,静置,离心,沉淀,用240mL蒸馏水洗两次,水洗第二遍时pH5~6,240mL 85%乙醇洗两次,离心,沉淀,60℃真空干燥,得黄芩提取物。
Ⅴ组:加浓HCl 4.6mL,调pH1~2,80℃水浴,恒温30min,沉淀,加120mL蒸馏水,搅匀(有黏液)难滤,加6.6mL 40% NaOH,调至pH7,加120mL 95%乙醇,不溶解,离心(4000r,20min)后,加浓HCl 7.8mL,调pH1.0~2.0,60℃水浴保温30min,静置,离心,沉淀,用700mL蒸馏水洗两次,水洗第二遍时pH5~6,700mL 85%乙醇洗两次,离心,沉淀,60℃真空干燥,得黄芩提取物。
Ⅵ组:加浓HCl 4.6mL,调pH1~2,80℃水浴,恒温30min,沉淀,加120mL蒸馏水,搅匀(有粘液)难滤,加6.6mL 40% NaOH,调至pH7,加120mL 95%乙醇,不溶解,离心(4000r,20min)后,加浓HCl 7.6mL,调pH1.0~2.0,60℃水浴保温30min,静置,离心,沉淀,用700mL蒸馏水洗两次,水洗第二遍时pH5~6,700mL 85%乙醇洗两次,离心,沉淀,60℃真空干燥,得黄芩提取物。
2.1.7 黄芩提取工艺验证 分别称取10份家种黄芩100g,加沸水煎煮,第1次12倍量,2h,第2次10倍量,1.5h,合并煎液,浓缩至7倍量(80℃,ρ1.00~1.05),用浓HCl调节pH至1.0~2.0,80℃保温30min,静置,滤过,沉淀物加3.5倍量水搅匀 ......
