响应面优化酶法提取平菇多糖工艺研究(1)

摘 要:目的:建立平菇多糖的含量测定方法,确定纤维素酶法提取平菇多糖的最佳工艺条件。方法:采用苯酚-硫酸法测定平菇多糖的含量。以平菇多糖得率为指标,考察酶解时间、酶解温度、pH值等3个因素对平菇多糖纤维素酶提取工艺的影响,采用响应面分析法优化工艺条件。结果:平菇多糖的最佳提取工艺条件为酶解时间135min、酶解温度49℃、pH5.30,在此条件下得率可达6.82%。结论 该法成本低、产品得率高,比传统水提法提高3.98%。

Abstract:Objective:To establish determination methods and optimal extraction conditions of polysaccharides from Pleurotus ostreatus treated with cellulase.Methods:Total carbohydrate content was determined by the phenol-sulfuric acid method and the yield of polysaccharides was the indice to determine extraction process by a combination of response surface methodology (RSM).the enzymolysis time,enzymolysis temperature and pH value were selected as factors and a three-factors-three-levels experiment was developed by Box-Benhnken central composite design method.Results:The optimal conditions of variables were determined as follows:enzymolysis time 139 min, enzymolysis temperature 49℃ and pH value 5.3.Under the condition above, the yield of polysaccharides was 6.82%.Conclusion:It was a ideal means with relative low cost and higher extraction efficiency of polysaccharides,which could be improved by 3.98 % through the treatment with cellulase than without.
