冠心合剂对家兔颈动脉球囊损伤后Ⅰ型胶原表达的影响(1)

     摘 要:目的:观察冠心合剂对兔颈动脉经皮冠状动脉腔内成形术(PTCA)损伤后细胞外基质Ⅰ型胶原蛋白表达的影响。方法:运用球囊原位扩张、拉伤致兔颈总动脉损伤建立PTCA术后内皮损伤模型。从术前3天开始各治疗组予相应剂量冠心合剂灌胃给药,分别于2、4、6周处死动物,免疫组织化学法检测I型胶原的表达。结果:各中药治疗组内膜胶原密度较模型组降低,差异有统计学意义(P<0.05);冠心合剂大剂量组6周的内膜胶原密度表达最少。结论:冠心合剂可保护兔颈动脉球囊损伤后内膜,其机制可能与减少血管壁胶原含量有关。

    Effect of Guanxin Decoction on Expression of Type-I collagen

    in Rabbits after Carotid Artery Balloon Injury

    Abstract:Objective:To observe the effect of Guanxin decoction on extracellular matrix collagen Ⅰexpression after percutaneous transluminal coronary angioplasty (PTCA) of rabbits. Methods:Endothelial injury models were established by using in-situ balloon expansion and strain-induced injury of rabbit carotid artery. The Guanxin decoction groups were fed correspondingly does 3 days before the operation. All the animals were sacrificed at the end of the 2ed, 4th and 6th weeks respectively and operated carotid arteries were obtained. Immunohistochemical Strept Avidin-Biotin complex (SABC) staining were used for observing arterial morphology and intimal collagen density. Results:The Guanxin decoction groups have lower levels of intimal hyperplasia and intimal collagen (P<0.05). The large-does Guanxin decoction group has the lowest densities of intimal collagen at the end of the 6th week. Conclusion:Guanxin decoction could decreased the neointimal hyperplasia after PTCA , the mechanism may relate to inhibiting the expressions of collagen typeⅠin the injured artery.

    经皮腔内冠状动脉成形术(PTCA) 已成为冠心病心肌血管再通的重要手段,但术后再狭窄已成为影响其疗效的主要因素。血管组织中胶原增生是引起病变的主要因素,其参与了再狭窄的形成过程【sup】[1]【/sup】。本研究通过建立在体兔颈动脉球囊损伤模型,采用免疫组织化学方法,观察冠心合剂对实验性动脉球囊损伤后Ⅰ型胶原蛋白表达的影响。

    1 实验材料与方法

    1.1 实验动物

    健康纯种雄性新西兰大白兔92只, 体重1.8~2.3kg,由浙江中医药大学动物中心提供。

    1.2 主要实验药品及试剂

    冠心合剂(黄芪30g,蒲黄20g,五灵脂20g),将各药加水煎煮2次,第1次1.5h,第2次0.5h,浓缩成2g/mL的生药煎剂(杭州市中医院制剂室制备)。戊巴比妥钠(国药集团化学试剂有限公司);青霉素钠(华北制药有限公司);SABC一抗、山羊抗兔IgG抗体-HRP多聚体(武汉博士德公司) ......
