脾虚证荷瘤小鼠模型的建立(1)

摘 要：目的：利用泻下加劳倦过度方式结合皮下接种H-22肝癌细胞的模式造成脾虚型荷瘤小鼠模型。方法：16只昆明小鼠随机分为对照组和模型组,每组8只，10天后取食量、体重、肛温、腹泻、肛门污秽、拉尾排便次数、悬空拉尾抵抗时间及一般状况(拱背、毛色枯槁及脱毛、倦卧嗜睡等)，作小鼠脾虚征象详情记录。取血测定血清D-木糖醇、血清胃泌素、处死剖取胸腺、脾脏称重，计算胸腺指数和脾脏指数,评价模型的建立和病理生理特点。结果：造模后，造模小鼠腋下均长出肿瘤结节，并且出现明显食少、稀便、肛门污秽、神软萎靡、喜聚堆倦伏、毛色枯槁、拱背竖毛、活动缓慢、反应迟钝等脾虚证状，以第6天最显著。结论：泻下加劳倦过度方式结合皮下接种H-22肝癌细胞的模式在小鼠体内可以成功建立病证结合的脾虚证荷瘤模型。

Abstract:Objective:With the method which is combined with purgation and excessive fatigue and the mode of hypodermic inoculation of H22 hepatic carcinoma cells to establish the tumor-bearing mice models with Spleen Deficiency.Method:A total of 16 mice were randomly divided into the control group and the model group, with eight in each group.10 days later, conditions of appetite，weight，rectal temperature，diarrhea，excrement around the anus，times of defecation when pulling the tails，time length of resistance when pulling the tails hang in the air and general conditions(arch back，withered and yellow coats，unhairing，lassitude and drowsiness and so on )were recorded in detail as the symptoms of spleen deficiency. 3Get blood for the Determination of serum D-xylitol and gastrin(GAS).And the mice were sacrificed, weigh the mice's thymuses and spleens and calculate the thymus index and spleen index, evaluate the establishment of the model and Pathophysiologic Characteristics. Results :4It showed that after establishing the models, each mouse in model group grows tumor nodules in axillary region. And accompanied with apparent spleen-deficiency symptoms such as poor appetite, loose stool, excrement around the anus, lassitude ,tend to rest in groups, withered and yellow coats, arch its back and piloerection, slow-moving, bad-response, especially marked at the sixth day.Conclusion: 5the method combined with purgation and excessive fatigue and the mode of hypodermic inoculation of H22 hepatic carcinoma cells can set up tumor-bearing spleen deficient mice models with combination of disease and syndrome.
