芍药软肝方对荷肝癌H22小鼠肿瘤组织TGF-β1RⅡ、NF-κB、VEGF表达的影响(1)

摘 要:目的:观察芍药软肝方对小鼠H22肝癌肿瘤组织TGF-β1RⅡ、NF-κB、VEGF基因表达的影响。方法:ICR荷 H22肝癌小鼠50只,随机分生理盐水对照组,芍药软肝方低、中、高剂量组,环磷酰胺组,停药次日处死小鼠,取皮下肿瘤组织。采用荧光定量RT-PCR法测定芍药软肝方对小鼠 H22肝癌组织TGF-β1RⅡ、NF-κB、VEGF基因的表达。结果:芍药软肝方能上调小鼠 H22肝癌组织TGF-β1RⅡ表达水平,以中剂量组最明显,与生理盐水组相比有显著性差异(P<0.01)。芍药软肝方各给药组均能明显下调荷H22肝癌实体瘤小鼠肿瘤组织中NF-κB的表达水平,与生理盐水组相比均有显著性差异(P<0.01)。芍药软肝方中、高剂量组能明显降低ICR小鼠H22肝癌肿瘤组织中VEGF的表达水平,以高剂量组最明显,中、高剂量组与生理盐水组均有显著差异(P<0.01)。结论:芍药软肝方抑瘤作用的机理可能是通过下调荷瘤小鼠肿瘤组织NF-κB、VEGF表达,并上调TGF-β1RⅡ的表达水平,从而抑制肿瘤组织新生血管生成,抑制肿瘤细胞增殖,实现其抗肿瘤作用。

Abstract:Objective:To investigate the effect of Shaoyao Ruangan Formula on the expressions of TGF-β1RⅡ,NF-κB and VEGF in mice bearing Hepatoma H22 tissues. Methods:Fifty ICR mice bearing Hepatoma H22 were divided into 5 groups:saline(NS)group,the low moderate and high dose with Shaoyao Ruangan Formula groups and Cyclophosphamide(CTX)group. The expressions of TGF-β1RⅡ,NF-κB and VEGF in tumor tissues were detected by quantitative real-time polymerase chain reaction(QRT-PCR). Results:Shaoyao Ruangan Formula could up-regulate the expression levels of TGF-β1RⅡ mRNA in tumor tissues from the mice bearing Hepatoma H22. Compared with saline control group,the effects of the mid dose groups were the most prominent and showed significantly difference in statistics(P<0.01). Shaoyao Ruangan Formula could down-regulate the expression levels of NF-κB mRNA in tumor tissues from the mice bearing Hepatoma H22. Compared with saline control group,the effects of the high dose groups were the most prominent and showed significantly difference in statistics(P<0.01).
