β-榄香烯联合高温对肺腺癌A549细胞作用的实验研究(1)

     摘 要:目的:观察榄香烯联合高温对人肺腺癌A549细胞的协同杀伤作用。方法:用MTT法检测高温处理或常温处理细胞对榄香烯的敏感性。采用15μg/mL榄香烯或60μg/mL榄香烯联合42℃高温处理1.5h或常温处理,使用流式细胞仪检测细胞周期,透射电镜观察细胞形态学改变。结果:高温作用后,榄香烯对A549细胞的IC50从35.43μg/mL降低至28.68μg/mL;MTT法检测显示低浓度榄香烯联合高温对A549细胞具有明显的协同杀伤作用,细胞呈现凋亡形态,可见凋亡小体,同时有细胞坏死。结论:低浓度榄香烯联合高温对A549细胞有协同细胞毒效应。

    Experiment Research on Synergistic Effect of Elemene Combined with

    Hyperthermia on Cell line A549 of Lung Adenocarcinoma

    Abstract:Objective:To observe the synergistic effect of elemene combined with hyperthermia on cell line A549 of human lung adenocarcinoma.Methods:The sensitivity of the cells treated with gradient concentration and elemene in 42℃ or 37℃ water bath for 1.5 hours were detected by MTT assay. One hour and a half after treatment with water bath at 42℃ combined with 15μg/mL or 60μg/mL of elemene,cell cycle was detected by flow cytometry and appearance of the cells was detected by transmission electron microscope. Results:After hyperthermia,IC50 values of elemene decreased from 35.43μg/mL to 28.68μg/mL. MTT assay showed low concentration elemene combined with hyperthermia had synergistic cell killing effect,and the cells of A549 appeared apoptosis. Apoptotic body and cellular necrosis can be seen. FCM showed elemene combined with hyperthermia had significant effect on cell cycle with increasing G0/G1 phase population and reducing S phase population. Conclusion:Low concentration elemene combined with hyperthermia has synergistic effect of cytotoxicity on A549 cells.

    近十年来肺癌的发病率和死亡率呈迅速上升的趋势。然而肺癌的治疗效果多年来一直没有显著提高,总的治愈率仅为10%左右。因此多学科综合治疗成为肺癌治疗的发展方向。热疗已经成为肿瘤的重要治疗手段之一,但容易诱发产生热耐受而影响疗效。β-榄香烯是从莪术中提取而来,它有直接的肿瘤抑制作用。一些研究表明,β-榄香烯可以提高一些肿瘤细胞对热疗的敏感性[1],为此,我们以人肺癌细胞株A549为对象,观察β-榄香烯联合高温对肿瘤细胞的毒性作用。
