肾脏间质树突状细胞在多器官功能障碍综合征中的变化与作用(1)
【摘要】目的 探讨肾脏间质树突状细胞在多器官功能障碍综合征免疫紊乱中的影响与作用。方法 腹腔注射酵母多糖制造C57BL/6小鼠MODS模型,分为正常、3~6 h、12~48 h、5~7 d和10~12 d组。观察各组小鼠肾脏间质树突状细胞的病理学变化及其表面标记物CD11c、CD205、CD80和I-Ab在MODS各期肾脏中的表达水平;流式细胞术检测外周血CD4+与CD8+的T细胞数量与比值。结果 在急性损伤期,肾脏间质树突状细胞大量增生,CD 80和I-Ab表达较正常组差异具有统计学意义(P+/CD8+比值则明显下降(Pb表达较急性损伤期显著减少(P+/CD8+比值下降至最低(Pb

【Abstract】Objective To explore the role of kidney interstitial dendritic cells in immunodissonance moechanism in mice with multiple organ dysfunction syndrome(MODS).Method The model of MODS wasmade by injecting zymosan into the peritoneal caiecty of C57BL/6 mice,and the micewere randomly divided into 5 groups,namely, normal,3~6 hours,12~48 hours,5~7 days,10~12 days after administrating zymosan.Pathological changes of kidney interstitial dendritic cells were observed by transmission electronic microscopes.Specific surface markers CD205,CD11c,CD80,MHCⅡ molecules I-Ab,CD4+ and CD8+ T lymphocyte subgroups in peripheral blood were detected by immunohistochemistry and flow cytomctry.Results In acute injury stage,in comparison with normal group,interstitial dendritic cells had a continuous proliferation with high expression of CD80 and I-Ab(P+/CD8+ declined markedly in peripheral blood(Pb declined to very low level(P+/CD8+ declined remarkably to the minimum in periphend blood(Pb

对于多器官功能障碍综合征(multiple organ dysfunction syndrome,MODS)发病机制的研究一直是危重病急救医学领域的热点问题。近年来,免疫系统功能紊乱在MODS的发病过程中的作用受到了广泛关注。树突状细胞(dendritic cell,DC)作为功能强大的专责性抗原提供细胞,是提供抗原及启动T细胞免疫应答反应的关键细胞。笔者以往研究表明,免疫器官胸腺和脾脏中的树突状细胞参与了MODS免疫损伤的启动机制,是引发MODS中免疫抑制的重要免疫细胞学因素[1-3]。但是,关于MODS脏器免疫微环境中的间质树突状细胞的相关研究在国内外鲜有报道。笔者运用小鼠MODS模型,探讨肾脏间质DC在免疫紊乱及脏器损伤机制中的变化与作用,为MODS免疫学机制与防治研究提供新的依据。
1 材料与方法
1.1 实验动物与模型
