高钾致死大鼠心脏热缺血后心脏结构和功能的变化(1)

摘要:目的研究高钾致死后大鼠不同热缺血后心脏结构和功能的变化,以判定供心的热缺血时间限制。方法32只Wistar大鼠随机分为对照组(A组,热缺血2 min)、热缺血5 min组(B组,热缺血5 min)、热缺血10 min组(C组,热缺血10 min)、热缺血15 min组(D组,热缺血15 min),每组8只。热缺血结束后建立Langendorff模型:观察心脏复苏情况,测定平均冠脉流量,描记左室内压力波形,计算相应血流动力学指标,进行组织学检查。结果A组、B组心脏复苏显著优于C组、D组,前两组在复灌初始即全部恢复窦性心律,C组有5只心脏在灌注10 min之内自动恢复窦性节律,偶有室早,另外3只初始为室颤,经心脏按压后2只复律,1只持续室颤至试验结束仍未复律。D组复灌初始全部为室颤,经灌注15 min~20 min后,5只心脏恢复窦性节律,另外3只经反复按摩至试验结束仍未复律。A、B、C、D 组血流动力学各项基础值差异有统计学意义,B组与A组比较无统计学意义(P>0.05),C组、D组结果明显低于A组和B组(P<0.01)。C组与D组各项指标差异无统计学意义(P>0.05)。结论高钾致死大鼠心脏常温热缺血10 min以上不宜用于心脏移植。

Heart Function and Myocardial Structure Following Heart Warm-ischemia Caused by Hyperpotassemia in Rats

Abstract:ObjectiveTo research the heart function and myocardial structure of non-beating heart warm ischemia caused by hyperpotassemia in rats in order to conform the warm-ischemia time limit of the donor heart.MethodsThirty-two Wistar rats were divided into 4 groups randomly:Group A (n=8,warm ischemia 2 min),group B(n=8,warm ischemia 5 min),group C (n=8,warm ischemia 10 min),group D(n=8,warm ischemia 15 min).After the warm ischemia,Langendorff Model was established.The resuscitation of the hearts was observed,and coronary flow was measured.The intra-left ventricular pressure was record and hemodynamic values were calculated.The samples of heart were observed by light microscope.ResultsThe resuscitation in group A and group B was better than that in group C and group D.The hearts in group A and B got sinus rhythms as soon as reperfusion initiated.While ventricular fibrillation (VF) was developed in all hearts in group D.Five out of eight hearts in group D had sinus rhythm 15 min to 20 min after reperfusion started,there were three VF rhythms with continuous compression in this group ......
