硝苯地平缓释胶囊体外释放度研究

摘要：目的建立硝苯地平24 h缓释胶囊的释放度研究方法。 方法采用紫外分光光度法，以硝苯地平缓释胶囊为样品，考察释放介质、装置及转速对其释放度的影响。结果2 h前以1% 十二烷基硫酸钠的0.1 mol/L盐酸900 mL为释放介质，2 h后以1%十二烷基硫酸钠的磷酸盐-枸橼酸缓冲液(pH 6.8)900 mL为释放介质，转篮法，转速为100 rpm，恒温(37±0.5)℃，缓释胶囊的释药行为良好。 结论该法可以作为硝苯地平24 h缓释胶囊的体外释放度研究方法。

Study on Release Rate of Nifedipine Sustained Release Capsule in Vitro

Abstract：Objective To study the release rate of nifedipine 24 h sustained release capsule.Methods The release rate of nifedipine 24 h sustained release capsule was assayed by ultraviolet spectrophotometric method. The dissolving medium, device and rotation speed were selected according to the release rate of the capsule. Results The release behaviors of nifedipine 24 h sustained release capsule were good in 900 mL 0.1 mol/L HCl solution with 1% SLS (medium A)in first two hours and in 900 mL phosphate-citric acid buffer solution (pH6.8) with 1% SLS(medium B) after two hours with the rotation speed of 100 rpm by basket stirring method at (37±0.5)℃.Conclusion This method can be used to determine the release rate of nifedipine 24 h sustained release capsule.

硝苯地平(nifedipine，NF)为二氢吡啶类钙通道阻断剂，具有舒张外周及冠状动脉的功能，临床上广泛用于高血压和冠心病[1]。但硝苯地平半衰期短，血药浓度波动大，可引起心血管方面的严重副反应[2]。为了减少副作用，减少用药次数，增加患者的顺应性，我们研制了硝苯地平24 h缓释胶囊。本文对自制硝苯地平24 h缓释胶囊的释放度进行了研究。
1材料和方法
1.1药品与试剂
NF(上海德胜集团安庆有限公司)；NF缓释胶囊(自制，30 mg/粒，批号050406，05052402，05052403，05052703)；聚乙烯吡咯烷酮 K30(PVP-K30，美国国际特品公司)；乙基纤维素 N-10(EC，上海卡乐康)，羟丙基甲基纤维素E5(HPMC-E5，上海卡乐康)；丙烯酸树脂 S100(Eudragit S100，德国罗姆)；丙烯酸树脂 L100(Eudragit L100，德国罗姆)；十二烷基硫酸钠(SLS，广州器化医疗设备有限公司，进口分装)；其它试剂均为分析纯。
1% SLS的0.1 mol/L盐酸溶液为介质A，1% SLS的磷酸盐-枸橼酸缓冲液为介质B。
1.2仪器
PHS-25型酸度计(上海伟业仪器厂)；流化沸腾制粒包衣机(瑞士Niro- Aerometic)；ZRS-8G智能溶出试验仪(天津大学无线电厂)； UV-1601紫外可见分光光度计(日本岛津) ......
