肝素寡糖对人外周血T细胞分泌白介素-4和白介素-5的影响

     摘要:目的研究肝素寡糖(Oligs)对人外周血T细胞(PBTLs)分泌白介素-4(IL-4)和白介素-5(IL-5)的抑制作用。方法用3种方法降解肝素并用凝胶过滤色谱法制备肝素寡糖。抽取10例过敏性嗜酸粒细胞性胃肠炎患者的PBTLs用植物凝集素(PHA)和Oligs处理,用ELISA法测定PBTLs培养的上清液分泌IL-4和IL-5的含量。 结果Oligs样品浓度5 μg/mL时,可显著抑制PBTLs对细胞因子IL-4、IL-5的分泌。但是,不同相对分子质量(Mr)的Oligs对IL-4和IL-5具有不同的作用。对IL-4抑制作用最强的是用过氧化氢降解肝素所得Mr为1 142的四糖。抑制IL-5作用最强的是用β-消除法降解肝素所得Mr为1 806的六糖。结论Oligs对人PBTLs分泌IL-4和IL-5的抑制作用与分子结构密切相关,并且有一个基本结构。对IL-4和IL-5抑制作用最强的分别是四糖和六糖。

    Effect of Heparin-Derived Oligosaccharides on Secretion of Interleukin-4 and Interleukin-5 from Human Peripheral Blood T Lymphocytes

    Abstract:Objective To study the inhibitory effect of heparin-derived oligosaccharides (Oligs) on the secretion of IL-4 and IL-5 from human peripheral blood T lymphocytes (PBTLs). Methods Heparin was degraded by three different methods and low molecular weight heparins (LMWHs) were purified with gel-filtration chromatography in order to prepare Oligs. The PBTLs from ten patients with allergic eosinophilic gastroenteritis were treated with phytohemagglutinin (PHA) and Oligs, and the concentrations of IL-4 and IL-5 in the cell culture supernatant fluid were determined by ELISA. Results 5 μg/mL of Oligs samples could obviously inhibit the secretion of IL-4 and IL-5 from human PBTLs, while Oligs with different molecular weights(Mr)had different effects on the secretion of IL-4 and IL-5. A tetrasaccharide with Mr of 1 142 obtained by H2O2 degradation could most inhibit the secretion of IL-4, while a hexasaccharide with Mr of 1 806 obtained by beta-elimination could inhibit the secretion of IL-5 most ......
