[摘要] 目的:建立HPLC-MS/MS法测定益智合剂中芍药苷含量的方法。方法:采用高效液相串联质谱法对益智合剂中痕量的芍药苷进行分析,选用ZORBAX C18反相柱(2.1 mm×30 mm,3.5 μm)为色谱柱,流动相为甲醇-5 mmol/L甲酸铵(75∶25),流速0.4 ml/min,柱温25℃。质谱条件为电喷雾离子源(ESI),检测方式为多离子反应监测(MRM),负离子模式,用于定量分析的离子为m/z 525→449。结果:用外标法定量分析,线性范围为20~5 000 μg/L,r=0.999 9;益智合剂中芍药苷的最低检测限为2 μg/L;精密度RSD为0.84%;平均加样回收率为96.34%,RSD为2.45%(n=5);测得益智合剂中芍药苷的含量为31.45 μg/L。结论:本法操作简单、快捷,结果准确可靠,可作为目前有效检测益智合剂中的痕量成分芍药苷的含量。

Determination of the content of paeoniflorin in Yizhi Mixture by HPLC-MS/MS

[Abstract] Objective: To establish a HPLC-MS/MS method for the determination of paeoniflorin in Yizhi Mixture. Methods: HPLC-MS/MS was used for he determination of paeoniflorin, ZORBAX C18 column(2.1 mm×30 mm,3.5 μm) was used with methanol-5 mmol/L ammonium formate solvent mixture solution(75∶25) as the mobil phase, the flow rate was 0.4 ml/min,column temperature was 25℃. Electrospray ionization source was used. Paeoniflorin (MRM m/z 525→449) were detected by the agilent 6 410 triple quad mass spectrometer system. Results: The linear range was 20-5 000 μg/L (r=0.999 9), the mean recovery was 96.34%, RSD=2.45%(n=5). Conclusion: The method is simple, sensitive and accurate.It can be used for the quality control of Yizhi Mixture.

益智合剂为脑科医院自行研制的合剂,主要用于儿童多动症的临床治疗[1]。该中药方剂主要由鹿角霜、山茱萸、浮小麦、石菖蒲、黄柏、远志、白芍等17味中药材组成,经加水煎煮2次,合并滤液浓缩而成。传统中医学认为该方具有增补肝肾精血,肝阳得潜,心神安宁,使多动、注意力涣散等症状消除等功效[1-3]。为有效控制该制剂质量的可控性、重现性和稳定性,保证临床用药安全有效,我们拟建立测定该合剂中芍药苷含量的方法。

HPLC Agilent 1 200,Agilent 6 410 Triple Quad LC/MS,Agilent色谱化学工作站(软件版本编号B.01.00);旋涡混合器(原上海医科大学仪器厂);电子天平(瑞士SARTORIUS);高速离心机(美国MICRO12-24);真空干燥箱(德国MMM-GROUP)。
