[摘要] 目的:建立HPLC法测定维生素B2的含量方法。方法:色谱柱为Agilent ZORBAX SB-C18(150 mm×4.6 mm,3.5 μm);流动相为甲醇-0.35%磷酸的0.01 mol/L庚烷磺酸钠溶液(26∶74);检测波长267 nm;流速1.0 ml/min;柱温35℃。结果:维生素B2在1.5~12 μg/ml范围内线性关系良好,回归方程为Y=94.617 65X-0.467 68(r=0.999 95,n=6),低、中、高三种浓度的回收率分别为99.48%、99.26%、100.54%,RSD分别为0.37%、0.18%、0.18%。结论:该法快速、准确,灵敏度高,重现性好,可用于维生素B2的质量控制。

Determination of vitamin B2 in vitamin B2 bulk drug with HPLC

[Abstract] Objective: To establish a HPLC method for the determination of content of vitamin B2 in vitamin B2 bulk drug. Methods: The Agilent ZORBAX SB-C18(150 mm×4.6 mm,3.5 μm)was used, mobile phase was methanol-0.35%phosphoric acid and 0.01 mol/L sodium heptanesulfonate solution (26∶74) with the flow rate of 1.0 ml/min, detected wavelength was set at 267 nm, column temperature was 35℃. Results: The calibration curve was linear at the range of 1.5-12 μg/ml for vitamin B2. The linear equation was Y=94.617 65X-0.467 68(r=0.999 95, n=6), The average recovery of vitamin B2 in the low, medium and high concentrations were 99.48%, 99.26% and 100.54%, respectively; RSD were 0.37%, 0.18%, 0.18%, respectively. Conclusion: This method is rapid, accurate, high sensitivity and good reproducibility, it can be used for quality control of vitamin B2 bulk drug.

维生素B2,又名核黄素,为水溶性维生素,容易消化和吸收。其功效为促进发育和细胞的再生,帮助消除口腔内、唇、舌的炎症以及增进视力,减轻眼睛的疲劳等。目前维生素B2的含量测定方法主要为紫外-可见分光光度法[1]和荧光分光光度法。本文用HPLC法测定维生素B2的含量[2-4],并与紫外-可见分光光度法进行比较,结果表明,HPLC法快速、准确、灵敏度高、重现性好。
1仪器与试药
1.1仪器
Agilent1200高效液相色谱仪(四元泵、柱温箱、VWD检测器、Agilent1200工作站);AG245电子分析天平;UV-2450型紫外-可见分光光度仪。
1.2试药
维生素B2对照品(中国生物制品检定所,批号:0718-200004);维生素B2原料药(北京蓝健兴达工贸有限公司提供);甲醇为色谱醇(美国Merck公司);水为超纯水,其他试剂为分析纯。
