[摘要] 目的:研究壬基酚(nonylphenol,NP)对大鼠子代睾丸卵泡刺激素受体(FSHR)mRNA表达的影响。方法:将40只雌性SD大鼠随机分为5组,NP低、中、高剂量组、阴性对照组和阳性对照组。NP各剂量组在妊娠第14~19天给予20、40、80 mg/(kg·d)壬基酚灌胃,阴性对照组给予同体积的花生油,阳性对照组给予30 μg/(kg·d)雌二醇,孕第19天取胎鼠睾丸,用实时荧光定量PCR检测睾丸中FSHR mRNA的表达。结果:与阴性对照组比较,低剂量组表达升高,但随着剂量的增加,FSHR mRNA表达下降,差异有统计学意义。结论:壬基酚可致大鼠子代生殖毒性,影响FSHR的转录水平,导致生精障碍。

Effects of nonylphenol on the expression of FSHR in rat embryonic testes

[Abstract] Objective: To study the effects of nonylphenol (NP) on the expression of FSHR mRNA in rat embryonic testes in vivo. Methods: 40 female SD rats were randomly divided into five groups, low, middle and high dose NP groups, negative control group and positive control group; and low, middle and high dose NP groups were intragastrically administered with 20, 40 and 80 mg/(kg·d) NP after 14 to 19 days of pregnancy, negative control group was administered with peanut oil with same volume, and positive control group was administered with 30 μg/(kg·d) Estrogen. Embryonic testes were collected at the nineteenth day after pregnancy. The expression of FSHR mRNA was determined by Real-time PCR. Results: The expression of FSHR mRNA in low dose group was increased compared with negative control group, but with the increase of NP dose, the expression of FSHR mRNA decreased, and it showed a significant difference. Conclusion: NP could result in the reproductive toxicity of rats, affects the transcriptional level of FSHR, and further results in spermatogenesis dysfunction.

壬基酚(nonylphenol,NP)是全球商用第二大类非离子表面活性剂壬基酚聚氧乙烯醚(NPES)的主要降解产物[1]。NP为具有雌激素活性的环境毒物[2],其雌激素效力为17β-Estradiol的1/(103~106)[3]。研究表明,NP可影响雄性动物的生殖系统,减少和降低精子生成的数量和质量。1992年Carlsen等[4]首先报道,根据过去50年的资料统计,人类精子计数下降了40%~50%;张树成等[5]报道,我国1981~1996年有生育能力的男性的精子计数、精子活动率、正常精子形态、精液量和排精总数呈下降趋势 ......
