[摘要] 目的:采用RP-HPLC法测定保健食品银杏叶软胶囊中总黄酮的含量。方法:采用Agilent C18(4.6 mm×250 mm,5 μm)色谱柱,流动相为甲醇-水(用磷酸调pH 为2.5 )(50∶50),流速为1.0 ml/min,检测波长为360 nm,柱温为30℃。结果:槲皮素、山奈素、异鼠李素的线性范围分别为0.050~0.400、0.005~0.040、0.005~0.040 μg;3批样品中总黄酮含量分别为6.466、6.455、6.838 mg/粒。结论:本法简便、准确、分离效果好,适合于银杏叶软胶囊总黄酮的含量测定。

Determination of Total Flavonoid in Yinxingye Soft Capsules by HPLC

[Abstract] Objective: To establish an RP-HPLC method for quantitative determination of Total Flavonoid in Yinxingye Soft Capsules. Methods: The analytical column was Agilent C18 (4.6 mm× 250 mm, 5 μm) column. The mobile phase was composed of methanol and water (with phosphoric acid pH=2) at a flow rate of 1.0 ml/min. The wavelength of UV detector was 360 nm and column temperature was 30℃. Results: The linear ranges of Quercetin, Kaempferol and Isorhamnetin were 0.050-0.400, 0.005-0.040, 0.005-0.040 μg, respectively. The contents of Total Flavonoid in three batches of samples were 1 Capsule 6.466, 6.455, 6.838 mg. Conclusion: This method is sensitive, repeatable and suitable to determine the content of Total Flavonoid in Yinxingye Soft Capsules.

银杏叶软胶囊是由银杏提取物与适量的基质制成的软胶囊,银杏提取物中含有黄酮类化合物和银杏总内酯两类生物活性物。其中,总黄酮类包括槲皮素、山奈素、异鼠李素等,共同作用于心脑和外周末梢血管系统,促进血液循环和改善末梢血液的运行,从而预防血管系统循环障碍所引起的各种症状。另外,黄酮类化合物有清除游离氧的抗氧化等药理作用[1]。本文用HPLC法测定总黄酮的含量,为建立银杏叶软胶囊质量控制方法提供依据。
1 仪器与试药
1.1 仪器
LC-2010高效液相色谱仪(日本岛津公司);KQ-300超声清洗仪(昆山市超声仪器有限公司);色谱柱Agilent C18(4.6 mm×250 mm,5 μm)。
1.2 试药
槲皮素对照品(供含量测定用,批号:10081-200406,中国药品生物制品检验所);山奈素对照品(供含量测定用,批号:110861-200303,中国药品生物制品检验所);异鼠李素对照品(供含量测定用,批号:110860-200205,中国药品生物制品检验所);甲醇为色谱纯,水为重蒸馏水,其他试剂均为分析纯,银杏叶软胶囊(批号:080510、080920、091026,广州美丽康有限公司)。
2 方法与结果
