华蟾素乳剂对小鼠Lewis肺癌的药效学研究(1)

[摘要] 目的：研究华蟾素乳剂对实体肿瘤的疗效及其可能的作用机制。方法：本研究运用Lewis肺癌小鼠模型，观察华蟾素乳剂对小鼠Lewis肺癌的疗效、免疫功能的影响。结果：华蟾素乳剂治疗小鼠Lewis肺癌能抑制肿瘤的生长，抑瘤率高达32.7%，稳定病灶，提高荷瘤小鼠的生存质量，明显提高校正廓清指数α(P<0.01)以及溶血素水平(P<0.05)，改善免疫功能。结论：华蟾素乳剂对小鼠Lewis肺癌有明显疗效，其作用机制与其本身对Lewis肺癌实体瘤有抑制作用和提高荷瘤小鼠免疫功能有关。

Experimental study of effects of Huachansu Parenteral Emulsion on Lewis lung carcinoma in mice

[Abstract] Objective: To study the therapeutic effects of Huachansu Parenteral Emulsion (HCSPE) in mice and possible mechanism. Methods: The mice with Lewis lung carcinoma was used as a model for observing the therapeutic effect, the immunomodulatory functions, and changes of quality of life. Results: HCSPE inhibited the growth of tumor as the tumor inhibition ratio of 32.7%, enhanced clearance index α (P<0.01) and serum hemolysin level (P<0.05) obviously, increased life quality and the immunity of mice with Lewis lung carcinoma. Conclusion: HCSPE has therapeutic effects in mice with Lewis lung carcinoma. It′s mechanism might be associated with improving the immunity of mice with Lewis lung carcinoma.

华蟾素是中华大蟾蜍皮的水制剂，临床发现它对消化系肿瘤有较好的疗效，有报道称华蟾素对人肝癌、胃癌、结肠癌细胞具有体外杀伤作用，但未明确其对肺癌的治疗效果[1-3]。目前，市售的华蟾素制剂以水溶性注射剂(HCSPE)为主，同时还有水溶性口服液、片剂及胶囊，但未见华蟾素乳剂的相关报道[4-6]。由于乳剂对药物具有缓释、靶向定位、稳定等作用以及降低毒性等独特优点，故近年来中药静脉注射乳剂在抗肿瘤、抗心血管、抗病毒等载体方面中的应用发挥了独特作用。将华蟾素制成乳剂[7]，利用蟾毒灵等蟾毒内脂类成分具有亲脂性的性质，可提高药物的稳定性和抗肿瘤有效成分的含量，降低药物用量；而且由于乳剂的缓释及靶向作用，也可增强药物的疗效，降低毒副作用。本实验运用Lewis肺癌小鼠模型，观察华蟾素乳剂对Lewis肺癌的治疗作用，根据碳粒廓清实验和小鼠溶血素实验结果探讨华蟾素乳剂的抗肿瘤作用机制，结果表明其抗瘤作用与其提高小鼠免疫功能有关。
1 材料与方法
1.1 药品
