[摘要] 目的:研究药对川芎-香附配伍对单味药材中指标成分的影响。方法:以川芎嗪、阿魏酸、藁本内酯和α-香附酮为指标成分,采用HPLC法分析川芎、香附及川芎-香附配伍前后变化。色谱条件:色谱柱为Inertsil ODS-3(4.6 mm×250 mm,5 μm);流动相为:甲醇-0.05%磷酸水溶液(梯度洗脱,0~20 min:45%~48%甲醇;20~25 min:48%~70%甲醇;25~50 min:70%~75%甲醇;50~55 min:75%~100%甲醇;55~60 min:100%~45%甲醇),检测波长为270 nm。结果:药对中指标成分川芎嗪、阿魏酸、藁本内酯含量与单味药材相比均有所增大。结论:川芎-香附配伍前后特征峰面积均发生明显变化。

Analysis of the diversify in the matched pair of Chuanxiong Rhizoma and
Cyperi Rhizoma by HPLC

[Abstract] Objective: To study the effect on the index component of Chuanxiong Rhizoma and Cyperi Rhizoma after matched. Methods: The ligustrazine, ferulic aid and ligustilide were used as the index ingredients to analyze the diversity in the matched pair of Chuanxiong Rhizoma and Cyperi Rhizoma by HPLC. The Inertsil ODS-3 (4.6 mm×250 mm, 5 μm) column was used; methanol-0.05% phosphoric acid was used as mobile phase in a gradient elution(0-20 min: 45%-48% methanol; 20-25 min: 48%-70% methanol; 25-50 min: 70%-75% methanol; 50-55 min: 75%-100% methanol; 55-60 min: 100%-45% methanol). The detective wavelength was 270 nm, and the flow rate was about 1 ml/min. Results: The concentration of ligustrazine, ferulic acid and ligustilide in crude drug pairs were increased compared with the single plant medicine. Conclusion: The peak area has changed absolutely after matched.

川芎-香附既是行气疏肝的药对,也是很多大复方中常用的配伍结构,临床主要用于郁证的治疗。川芎味辛,性温。入肝、胆、心包经,具有活血行气、祛风止痛的功效[1],为治头痛之要药[2]。香附味辛、微苦、微甘,性平,归肝、三焦经,具有疏肝行气、调经止痛的功效,常用醋制。
由于香附、川芎化学成分复杂,既含有大量的脂溶性成分,又有水溶性成分。本试验选择川芎中成分明确的生物碱主成分川芎嗪、酸性主要成分阿魏酸及挥发油主要成分藁本内酯;选择香附中成分明确的挥发油主成分α-香附酮作为指标,采用HPLC法分析药对川芎-香附配伍前后的含量变化,探讨川芎-香附配伍对指标成分的影响。
