[摘要] 目的:建立高效液相色谱法测定复合鱼肝油制剂中维生素A含量的方法。方法:采用高效液相色谱法,选择迪马(Diamonsil)C18(4.6 mm×250 mm,5 μm)色谱柱,流动相为:甲醇-水(95∶5),流速1.0 ml/min,检测波长为326 nm,柱温为 35℃。结果:维生素A进样量在2.907 0~34.883 7 IU/ml范围内呈良好的线性关系(r=0.999 6),平均回收率为98.84%。三个批次的复方鱼肝油制剂的含量分别为62.082、62.163、62.558 mg/瓶。结论:以高效液相色谱法检测复合鱼肝油制剂中维生素A的含量,结果准确、重现性好,精密度高,可作为该制剂的质量控制方法。

Determination of Vitamin A in Compound Morrhuate Preparation by HPLC

[Abstract] Objective: To develop an HPLC method for the determination of Vitamin A in Compound Morrhuate Preparation. Methods: The method was performed on Diamonsil C18(4.6 mm×250 mm, 5 μm) column. The mobile phase consisted of methanol-water (95∶5), at a flow rate of 1.0 ml/min. The UV detection wavelength was 326 nm and the injection volunm was 20 μl. The column temperature was 35℃. Results: The linear range of reserpine was 2.907 0-34.883 7 IU/ml (r=0.999 6), with average recovery of 98.84%. The average contents of the three batches Compound Morrhuate Preparation were 62.082, 62.163, 62.558 mg/bottle respectively. Conclusion: Determination of Vitamin A in Compound Morrhuate Preparation by HPLC is rapid ,accurate and suitable for the quality control of Compound Morrhuate Preparation.

维生素A(Vit A)是最早被认识并应用于临床的维生素,属脂溶性维生素,为全反式视黄醇的一组有活性的β2紫香酮的衍生物,包括视黄醇、视黄醛、视黄酸(RA)及其酯类[1]。Vit A对机体的生长发育、组织细胞增殖与分化均具有广泛的调控作用[2]。鱼肝油收载于《中国药典》2005年版,是维生素类非处方药,其中含有维生素A和D是人体生长发育的必需物质,尤其是对胎儿、婴幼儿的发育,上皮组织的完整性,视力,血钙和磷的恒定,骨骼、牙的发育等有重要作用[3]。复合鱼肝油制剂是由鱼肝油及多种维生素和辅料加工制成的溶液剂,临床用于预防和治疗因维生素A和D以及B族维生素缺乏所致的各种疾病。为了解本品质量,保证临床疗效,我科采用高效液相色谱法测定复合鱼肝油制剂中维生素A的含量,现报道如下:
1 仪器与试药
岛津LC-10ATVP高效液相色谱仪,大连中北色谱工作站;AT-201电子分析天平(分辨率d=0.01 mg,瑞士梅特勒),岛津UV-2550紫外-可见分光光度计。维生素A醋酸酯对照品(规格:20 mg,含量为100%,广州珠江制药厂提供);复合鱼肝油制剂(鱼肝油乳剂,规格:500 ml/瓶,国药准字H44024528,广州珠江制药厂生产,批号:081105;081102;080202)。甲醇为色谱纯,水为双蒸纯化水,其他试剂为分析纯。
2 方法与结果
