[摘要] 目的:采用高效液相色谱法,利用正交试验L9(34)探究野木瓜中有机酸的最佳提取工艺条件。方法:运用正交试验对贵州野木瓜干燥根茎的提取条件进行探究,采用HPLC测定反丁烯二酸的含量。结果:极差分析表明,在各提取条件中,对反丁烯二酸的含量影响的顺序是:溶剂种类>提取次数>提取时间>料液体积比;方差分析表明,溶剂种类的差异性大于其他三个因素。结论:运用正交试验得到野木瓜最佳提取工艺是:90%的乙醇提取3次,每次2 h,料液体积比8倍量较为合适,且各项考察结果均符合要求,提示该提取工艺切实可行,为野木瓜中反丁烯二酸的含量测定提供了实验依据。

Optimization of the extraction technics for fumaric acids from Starntonia chinensis DC. based on orthogonal test

[Abstract] Objective: To explore the best extraction conditions of fumaric acid in Starntonia chinensis DC. by using HPLC and orthogonal test L9(34). Methods: Orthogonal test was applied to probe the extraction process on dried leaf and stem of Starntonia chinensis DC. from Guizhou; HPLC was utilized to measure the fumaric acid content. Results: Range analysis showed that the extraction conditions on the content of fumaric acid was in the order: kinds of solvent > extraction times > extraction time > volume ratio of solid; variance analysis indicated that kinds of solvent had greater effects than other factors. Conclusion: The optimal condition of the orthogonal test is that, the solid dissolved in 90% ethanol, extraction lasted for 2 hours, 3 times, solid to liquid volume ratio of 8 is more appropriate and the results of method investigation comply with all requirements. The technique proves to be practical, which provides theoretical foundation for measuring the content of fumaric acid in Starntonia chinensis DC..

野木瓜(starntonia chinensis DC.),常用名七叶莲,为木通科(Lardizabalaceae)野木瓜属植物,性温,味微苦,具有祛风止痛及舒筋活络等功效[1-2]。临床用于各种手术止痛、坐骨神经痛、腰椎骨质增生、跌打损伤等,疗效显著,且具有无耐药性、无成瘾性、毒副作用小的特点[3-4],是卫生部公布的药食两用植物,全株具有较高的药用价值。原标准中有一个显微鉴别,但未见该药材质量标准研究的报道。为了探究野木瓜中反丁烯二酸的含量,本实验运用高效液相色谱法测定野木瓜中反丁烯二酸的含量,并进行方法学研究,为野木瓜的质量标准研究提供一定的实验数据。
