[摘要] 目的:研究黄连不同炮制品水煎剂中主成分含量的变化,探讨不同炮制方法对黄连抗菌活性的影响。方法:以小檗碱为黄连的代表成分,运用HPLC法测定4种不同黄连炮制品水煎剂中代表成分的含量。将黄连不同炮制品水煎液置于培养基中与细菌同时温育,观察药物对细菌生长繁殖的影响,以此判断药物抑菌作用的强弱。结果:黄连不同炮制品中小檗碱的含量明显不同,以药物最低抑菌浓度(MIC)显示黄连不同炮制品均有抑菌作用,但作用强弱不同。结论:不同炮制法不仅改变黄连药材的组分和含量,还可能通过影响其抗菌活性,使其药性及功能主治发生变化。

Influences of different processing methods on the content and the bacteriostasis effect in major constituent of Coptis chinensis

[Abstract] Objective: To investigate the content of major constituents in processed Coptis chinensis extracted by water, and to study the impact of different processing methods on the bacteriostasis effect. Methods: Berberine was chosen as representative constituent, of which the content in three differently processed Coptis chinensis extracted by water were determined by HPLC. Different processed products of Coptis chinensis in culture medium were put and cultivated with bacteria in the same time, and the influences of drugs on bacteria growth to judge the strength of bacteriostasis were observed. Results: The content of Berberine in differently processed Coptis chinensis were significantly different. It showed the bacteriostasis of different products all had the bacteriostatic action with MIC but in different strength. Conclusion: The content of major constituents and the bacteriostasis effect is changed by different means, different processing methods may transform the properties and functions of Coptis chinensis.

黄连是临床常用的药材,据来源不同分为“味连”、“雅连”、“云连”等,主产于四川、云南、湖北等地。其始载于《神农本草经》,列为上品,具有清热燥湿、泻火解毒之功效。现代研究表明其有抗菌作用,且抗菌谱广,常用于解热镇痛、抗肠道细菌感染。
经不同方法炮制黄连后,其成分会发生不同程度的变化,可导致药性及功能主治的改变[1-2]。黄连的炮制种类繁多,其中酒黄连、姜黄连、萸黄连一直沿用至今,亦为药典收录。有学者以甲醇、乙醇等有机溶剂为提取溶剂对黄连不同炮制品提取物进行研究[3],结合临床实际均是采用水煎剂入药。因此,本文选择黄连中小檗碱为主要有效成分代表,检测黄连不同炮制品水煎剂中小檗碱主要含量的变化[4-5]。不同炮制品的功效差异除与其有效成分有关,更与其抗菌活性密切相关[6]。本文研究了黄连不同炮制品中主要成分盐酸小檗碱含量的变化,及其对常见菌株抗菌活性的影响,为其临床应用提供参考依据。

高效液相色谱仪(Agilent 1100,美国安捷伦公司);超声波清洗器(KQ 3000,昆山市超声波仪器厂);电子分析天平(BP211D,德国赛多利斯);粉碎机(FW200,瑞安市永历制药机械有限公司)。
